大黄素对大鼠混合型高脂血症的影响

摘    要:目的研究大黄素对大鼠混合型高脂血症防治作用及可能的机制。方法健康雄性SD大鼠90只,随机分为6组(n=15):正常组,模型组、血脂康组(200mg·kg^-1)、大黄素高剂量组(300mg·kg^-1)、大黄素中剂量组(150mg·kg^-1)、大黄素低剂量组(75mg·kg^-1),除正常组外,其余各组每日灌胃高脂乳剂1次,共造模10wk,自第7周开始灌胃给药,每日1次,连续4周。腹主动脉取血,测定血清TG、TC、SOD、GSH—PX、MDA水平,取肝组织匀浆,测定TG、TC水平。结果大黄素可降低高脂血症大鼠血清TG(P〈0.05)和血清TC(P〈0.05);减少肝匀浆TG(P〈0.01)和肝匀浆TC(P〈0.05),降低MDA水平(P〈0.05),提高SOD、GSH—PX水平(P〈0.05)。结论大黄素有改善大鼠高脂血症大鼠血脂紊乱的作用,并不导致脂质在肝脏中蓄积,其作用机制可能与其抗氧化能力有关.

Abstract:Aim To observe the effects of emodin on combined hyperlipidemia familial in model SD rats and to explore its mechanisms. Methods Animal models with combined hyperlipidemia familial were established bv feeding high caloric diet for 10 weeks. The blood Iipid,MDA,SOD,GSH-PX,and liver t lipid were determined after therapy with emodin for 4 weeks. After 6 weeks,each drug group,Xu- ezhikang group(200 mg· kg-1 ) ,emodin group of high dosage(300 mg· kg-1) ,emodin group of intermediat dosage( 150 mg· kg-1 ) , emodln group of low dosage(75 mg· kg-1) were not only given high fat emulsion but also given corresponding drug. Result The animao model of combined hyperlipidemia familial were sucessfully made. Compared with the model group,the blood and liver lipid in treatment groups was signifcantly lowered( P 〈 0.05 ) ,the blood MDA was signifcantly lowered( P 〈 0.05 ) ,and the blood SOD and GSH-PX were significantly improved. Conclusion It is suggested that omodin might be effective in the prevention and treament of combined hyperlipidemia familial. Its functional mechanisms might lie in increasing the antioxidation of blood SOD, GSH-PX, and reducing blood MDA.
