摘    要:目的 研究槲皮素对卵巢癌细胞株HO 8910PM的体外侵袭能力的影响 ,并探讨其可能的作用机制。方法 卵巢癌细胞HO 8910PM经槲皮素处理后用Boyden小室的方法检测其体外粘附、侵袭和运动能力。明胶酶谱方法检测HO 8910PM细胞分泌的明胶酶活性。RT PCR方法检测TIMP 1、NM2 3H1基因的mRNA表达情况 ,用蛋白印迹方法检测c Fos,c Jun ,ERK2的蛋白表达变化。结果 卵巢癌细胞HO 8910PM经槲皮素处理后 ,体外侵袭、运动能力呈剂量依赖性下降 ,HO 8910PM细胞MMP 9的分泌下降。ERK2蛋白表达明显降低 ,但对TIMP 1、NM2 3基因的mRNA水平及c Fos ,c Jun蛋白表达无影响。结论 槲皮素在体外剂量依赖性地抑制卵巢癌细胞HO 8910PM的转移能力 ,这可能与槲皮素抑制MMP 9的分泌及下调ERK2蛋白表达有关。

Abstract:Aim To study the effect of quercetin on in vitro invasive activity of ovarian carcinoma HO 8910PM cell line, and to further elucidate the underlying mechanisms. Methods The in vitro invasion and mobility of ovarian carcinoma cell were measured in vitro by Boyden chamber assay after the treatment of qucertin. The gelatinase activity was tested by gelatin zymography assay. The expression of tissue inhibitor of metaloproteinase 1 (TIMP 1) and NM23H1 in mRNA level and that of c Fos, c Jun, ERK2 in protein level were measured by RT PCR and western blotting respectively. Results Quercetin significantly inhibited in vitro invasion and mobility of HO 8910PM cells. Quercetin inhibited the MMP 9 secretion from the HO 8910PM ovarian carcinoma cells, and dramatically decreased Erk2 expression in protein level, but qucertin neither affected expression of TIMP 1 and NM23H1in mRNA level, nor affected the expression of c Fos and c Jun in protein level. Conclusion Quercetin inhibits in vitro metastasis of highly metastatic ovarian carcinoma HO 8910PM cells in dose dependent manner. Inhibition of MMP 9 secretion by tumor cells is one of the possible mechanisms. Quercetin can decrease the expression of Erk2, which may be involved in the process of metastasis. Putting together, inhibition of MMP 9 secretion and ERK2 expression may play vital roles in the anti metastatic characteristics of quercetin.
