染料木素、芹菜素、槲皮素、芦丁和落新妇苷对高尿酸血症小鼠黄嘌呤氧化酶活性及血清尿酸水平的影响

摘    要:目的探讨黄酮类化合物染料木素、芹菜素、槲皮素、芦丁和落新妇苷体外对黄嘌呤氧化酶活性的影响,对高尿酸血症小鼠血清和肝脏黄嘌呤氧化酶活性的影响,同时评价对小鼠血清尿酸水平的作用。方法采用改良的紫外分光光度法测定染料木素、芹菜素、槲皮素、芦丁和落新妇苷体外对黄嘌呤氧化酶的抑制作用;采用尿酸酶抑制剂氧嗪酸钾诱导小鼠高尿酸血症模型,以分光光度法研究对小鼠血清和肝脏黄嘌呤氧化酶活性的影响,以磷钨酸法测定对小鼠血清尿酸水平的作用。结果体外实验表明这些黄酮类化合物对黄嘌呤氧化酶活性无明显影响。然而,体内实验观察到能够明显升高或降低黄嘌呤氧化酶的活性,而且,血清尿酸水平与血清黄嘌呤氧化酶活性密切相关,与肝脏黄嘌呤氧化酶活性无明显关联。该研究表明用这些黄酮类化合物给药的小鼠血清尿酸水平都高于正常对照组。结论这5种黄酮类化合物不能够作为替代别嘌醇的药物用来降低血清尿酸水平。
