黄芩苷对羟自由基诱发心肌损伤的保护作用

摘    要:目的:研究黄芩苷(baicalin,Bai)对羟自由基诱导大鼠心肌损伤的保护作用。方法:体外羟自由基发生系诱导心肌脂质过氧化,无机磷法测定心肌三磷酸腺苷(ATP)酶活性,分光光度法检测脂质过氧化物(MDA)含量、线粒体膨胀度和细胞色素氧化酶活性。结果:在羟自由基作用下心肌细胞膜中MDA含量明显升高,ATP酶活性显著降低,且ATP酶活性与MDA含量间呈显著负相关;线粒体膨胀明显,细胞色素氧化酶活性下降。Bai可降低保护羟自由基引起的心肌细胞膜MDA含量的升高和提高ATP酶活性;降低线粒体膨胀程度,恢复细胞色素氧化酶活性。结论:Bai可能通过清除氧自由基,抑制MDA的生成实现其保护心肌的功能。

Abstract:OBJECTIVE To study the protective effect of baicalin(Bai) on myocardial injury induced by hydroxyl free radical in rats.METHODS The hydroxyl free radical generation system was established by H_(2)O_(2)/Fe~(2 ) to induce myocardial lipoxidation.ATPase activity in membrane was determined according to the method of inorganic phosphate;Lipid peroxidation,mitochondria swelling and cytochrome oxidase activity were measured by spectrophotometry.RESULTS The increase of MDA content,decrease of ATPase,cytochrome oxidase activities,and mitochondria swelling by hydroxyl free radical were found in rat myocardium.A definite negative correlation between MDA content and ATPase activitiy was observed.Pretreatment with Bai was shown to have a significantly resisting effect on the changes of MDA content,ATPase and cytochrome oxidase activities,and mitochondria swelling induced by oxygen free radical.CONCLUSION Bai can protect myocardial injury by reducing MDA formation and scavenging hydroxyl free radical.
