江淮地区春柴胡HPLC特征图谱研究

摘    要:目的建立春柴胡HPLC特征图谱,为春柴胡的质量评价提供依据。方法色谱条件:Waters Symmetry C18色谱柱(250mm×4.6mm,5μm);流动相:乙腈-1mL·L-1甲酸,梯度洗脱;流速为1.0mL·min-1;检测波长为365nm。以"中药色谱特征图谱相似度评价系统2004A版"对数据进行分析。结果建立了春柴胡HPLC特征指纹图谱的共有模式,共标定了7个共有峰,指认了其中3个共有峰分别为槲皮素-3-O-葡萄糖醛酸苷、槲皮素和异鼠李素,并对10批春柴胡特征图谱与对照特征图谱计算相似度,其结果均大于0.96。结论建立的春柴胡HPLC特征图谱特征性和专属性强,方法快速、简便、可靠,可用于春柴胡药材的质量控制。

Abstract:Objective To establish an HPLC fingerprint of Bupleurum scorzonerifolium for quality control.Methods The chromatographic separation was performed on Waters Symmetry C18 column (250 mm×4.6 mm,5 μm) and mobile phase consisting of acetonitrile and 1 mL·L-1 formic acid in gradient elution.The flow rate was 1.0 mL·min-1 and the detection wavelength was 365 nm.Similarity evaluation system for Chromatographic Fingerprint of Traditional Chinese Medicine (2004A) was used in the analysis.Results The chromatographic fingerprint was completed with 7 recognizable peaks and 3 of them (quercetin-3-O-glucuronide, quercetin and isorhamnetin) were determined.The similarities of 10 batches of Bupleurum scorzonerifolium were higher than 0.96.Conclusion The established method could rapidly attain the identification and quality evaluation of Bupleurum scorzonerifolium.
