摘    要:目的:从成年大鼠纹状体区和胎鼠皮层分离并鉴定神经干细胞。方法:利用无血甭培养和单细胞克隆技术在成年大鼠纹状体区和胎鼠皮层分离具有单细胞克隆能力的细胞群,选取单个克隆进行连续传代培养获得大量亚克隆,采用免疫荧光细胞化学技术检测克隆细胞Nestin抗原和分化后特异性成熟神经细胞抗原的表达。结果:从纹状体区和皮层分离的细胞群具有连续克隆能力,胚胎早期细胞抗原-Nestin抗原阳性,分化后的细胞表达神经元

Abstract:Objective: To isolate the neural stem cells from the striatum of adult rats and the cortex of rat embryoes. Methods:Neural cells, which are able to form single cell cloning, were isolated from the striatum of adult rats and the cortex of rat embryoes with serum free incubation and single cell cloning technique. A single clone was incubated in consecutive generations in order to obtain large numbers of subclones. Immunofluorescence technique was employed to detect Nestin antigen and specific antigen of mature neural cells after the clone was differentiated. Results: The neural cells isolated from the striatum of adult rats and the cortex of rat embryoes showed the expression of Nestin antigen and were able to differentiate into neurons, astrocytes and oligodendrocytes. Conclusion: Our cell line belongs to neural precursor and is multipotent, which has the ability to undergo self renew. So, they are the stem cells of the central nervous system.
