丹参神经保护作用机制的研究

摘    要:目的 观察大鼠全脑缺血再灌注后凋亡相关基因bcl- 2蛋白表达情况以及丹参 (RSM)对其影响。方法 应用颈动脉负压分流法制备大鼠全脑缺血再灌注模型 ,采用免疫组织化学法检测全脑缺血再灌注后及经颈动脉注射丹参再灌注后不同时间大脑皮质及海马bcl- 2蛋白的表达情况。结果 大鼠全脑缺血 30min再灌注3h ,大脑皮质及海马bcl- 2蛋白表达至高峰 ;再灌注 6h其表达呈下降趋势。丹参干预组较对照组bcl- 2蛋白表达显著增加 (P <0 .0 1)。结论 bcl- 2主要通过抑制细胞凋亡的早期环节而发挥作用 ,丹参上调bcl- 2蛋白的表达进而发挥其神经保护作用

Abstract:Objective To study the effects of radix salviae miltiorrzhizae (RSM) on brain ischemic injury by examining the expression of bcl-2 protein. Methods The model of global cerebral ischemia/reperfusion was established by carotid negative pressure shunt in rats. RSM was administered into the (right) carotid artery at the beginning of reperfusion. Immunohistochemical method was used to detect the expression of bcl-2 protein in the cortex and hippocampus after different periods of reperfusion. Results The expression of bcl-2 in the course of reperfusion reached its peak level at 3 h and began to decline at 6 h, and the expression of bcl-2 protein was increased significantly in the test group as compared with that in the control group (P<0.01). However, no bcl-2 positive cells at all were noticed in the sham-operation normal control group. Conclusion Our results indicate that RSM may alleviate ischemia/reperfusion damage by reducing the apoptosis through its upregulation of bcl-2 protein expression.
