当归补血汤抗家兔动脉粥样硬化形成的实验研究

摘    要:目的观察中药当归补血汤在实验性动脉粥样硬化形成中降低血脂、减轻脂质过氧化损伤及减轻动脉粥样斑块形成作用.方法将新西兰兔50只随机分空白组、模型组、低剂量组、中剂量组和高剂量组,采用高脂饲料,同时用一次性静脉注射牛血清白蛋白的方法复制兔AS模型;以硫代巴比妥酸(TBA)比色法测定血清MDA,以羟胺比色法测定血清超氧化物歧化酶(SOD)、血清总胆固醇(TC)、血清甘油三酯(TG)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL);同时对主动脉和冠状动脉进行病理学检查.结果当归补血汤可明显地降低实验动物TC、LDL及丙二醛(MDA)的含量,升高SOD活性,并可使主动脉粥样硬化斑块面积及厚度明显小于模型组.结论当归补血汤调节血脂和抗氧化作用可能是其抗动脉粥样硬化作用的主要机制.

Abstract:Objective To observe the effects of Dangguibuxue Decoction on decrease of blood lipid, lipid peroxidation and atheromatous plaque in experimental atherosclerosis formation. Methods To divide randomly 50 rabbits into the blank group, model group, low-dose group, mid-dose group and high-dose group. To establish the rabbit atherosclerosis model by feeding fat food and intravenous injection of ox seralbumin. The level of serum malondialdehyde (MDA) was determined with thiobarbituric acid (TBA) chromatoptometry, and the levels of serum superoxide dismutase (SOD), total cholesterol (TC), triglyceride (TG), low-density lipoprotein (LDL) and high-density lipoprotein (HDL) were determined with hydroxylamine chromatoptometry in auto-biochemical analyzer of OLMPUS-A U600. At the same time the pathological examination was made on aorta and coronary artery. Results Dangguibuxue Decoction decreased obviously the levels of serum TC, LDL and MDA, increased the activity of SOD, and decreased the area and thickness of atheromatous plaque in experimental animals. Conclusion The antiatherosis mechanism of Dangguibuxue Decoction maybe related to its blood lipid regulation and antioxidation.
