摘    要:目的 探讨bcl-2反义寡核苷酸(antisense phosphorothioate oligodeoxynucleotide,ASODN)对胃癌SGC-7901细胞端粒酶活性的影响。方法 应用脂质体瞬时转染法介导bcl-2反义寡核苷酸,处理人胃癌细胞系SGC-7901后,分别采用Northern blot、Western blot检测bcl-2 mRNA、蛋白表达,用端粒微孔板杂交法检测端粒酶活性。结果 bcl-2 ASODN处理的胃癌SGC-7901细胞端粒酶活性明显受到抑制,并与ASODN的浓度和处理时间有关。结论 bcl-2反义寡核苷酸可抑制胃癌细胞端粒酶活性。

Effects of Antisense Phosphorothioate Oligodeoxynucletides Targeted againt bcl-2 mRNA on Telomerase Activity of Gastric Cancer SGC-7901 Cell
Abstract:Purpose To study the effects of bcl 2 antisense phosphorothioate oligodeoxynucleotide,(ASODN) on telomerase activity of gastric cancer SGC?7901 cell. Methods After gastric cancer SGC?7901 cell were treated by bcl 2 ASODN,we studies the expression of bcl 2 gene using Northern blot and Western blot,and telomerase activity using telomeric repeat amplification protocol(TRAP)?microtiter plate hybridization assay. Results bcl 2 ASODN can repress telomerase activity of gastric cancer SGC?7901 cell,and the effects of repression were related to treated time and dose. Conclusions bcl 2 ASODN can repress telomerase activity of gastric cancer cell.
