摘    要:目的探讨耳蜗毛细胞的分离方法。方法选出生3周的SD大白鼠,雌雄兼用,在自制的外淋巴液中取出耳蜗基底膜,置于0.25mg/ml木瓜蛋白酶中,37℃30min消化后,洗去酶液,用钢针在普通解剖镜下剥离毛细胞并置倒置相差显微镜下观察,记录成活时间。结果每侧耳蜗可获200左右成活的毛细胞,5h后仍有70%的毛细胞成活。结论用自制改良外淋巴液,并简化了操作过程,可以满意地获得形态、功能完好的毛细胞

Abstract:PURPOSE To develope a modified method of separating hair cells from mice. METHODS In the modified perilymph solution, the organ of corti was briefly incubated with papain and the hair cells were isolated from the tissue by mechanomanipulation under anotomy microscope. RESULTS Two hundred hair cells including IHC (inner hair cell) and OHC (outer hair cell) were obtained from each cochlea, and (with short term) approximately 70% of the cells retained good viability after 5 hrs' culture at room temperature. CONCLUSIONS Using the modified method the hair cells with good viability can be easily obtained.
