摘    要:目的 观察突变的大肠癌细胞线粒体DNA(mtDNA)转染 NIH3T3 细胞后转染细胞mtDNA D-环突变特性。方法 通过脂质体法( Lipofection2000TM) 将大肠癌细胞突变的 mtDNA 真核表达载体转染 NIH3T3细胞,利用 G418 抗性筛选克隆细胞;用PCR法检测转染细胞线粒体突变情况。结果 突变mtDNA 导致转染细胞 mtDNA 的突变和多态性变化。 结论 突变的大肠癌 mtDNA可致NIH3T3细胞mtDNA位点变化,但具体的机制和过程有待于进一步研究。

Abstract:【Abstract】Objective To observe the mtDNA mutation features of NIH3T3 cell after infected by mutated mtDNA from colorectal cancer cell line. Methods Recombinant eukaryotic cell expression plasmid of the mutated mtDNA was transfected into NIH3T3 cell using lipofection2000TM, and cloned cells were screened by G418. The mutation of transfected NIH3T3 cells mtDNA was detected by PCR. Results Mutation or polymorphism had been observed on the affection of mutated mtDNA NIH3T3 cell. Conclusion MtDNA mutations in colorectal cancer cell can affect the changes in NIH3T3 cell mtDNA sites, but the specific mechanisms and processes need be further studied.
