尾静脉注射STZ制备1型糖尿病大鼠模型实验的技巧

摘    要:目的探讨STZ尾静脉不同给药方法和补造模时不同给药剂量致大鼠糖尿病成模状况的差异。方法本实验通过尾静脉不同的给药方法(非盐水导入组、盐水导入组)以建立大鼠糖尿病模型。以及未成模的大鼠给予不同剂量的STZ补造模(全量组、2/3全量组)。观察给药后大鼠成模情况、一般状况及检测各组成模后3d、4W、8W血糖值的变化。结果盐水导入方法可以大幅度降低STZ尾静脉注射造模时的死亡率(6.52%),并提高成模率(73.9%)。补造模时2/3剂量组死亡率降低(6.67%,P〈0.05)且成模后血糖各组间,各时间点无差异(P〉0.05)。结论盐水导入的尾静脉给药方法是注射链脲佐菌素致大鼠糖尿病模型的最佳途径,补造模时以2/3剂量为宜。
