健脾清化方对2型糖尿病大鼠肝脏糖异生的影响

摘    要:目的:探讨健脾清化方对2型糖尿病大鼠肝脏糖异生的影响。方法:取Wistar大鼠,除正常组外,采用高脂喂养联合小剂量链脲左菌素(STZ)腹腔注射诱导2型糖尿病大鼠模型,成模后随机分为模型组、健脾清化方组。健脾清化方组予以含生药15 g·kg~(-1)·d~(-1)的混悬液灌胃,正常组和模型组用同体积双蒸水灌胃。连续灌胃4周后观察各组大鼠葡萄糖耐量和丙酮酸耐量,Western blot法测大鼠肝脏糖异生相关蛋白pFOXO1、FOXO1、PEPCK、G6Pase蛋白的表达,RT-PCR法测大鼠肝脏PEPCK、G6Pase的mRNA表达。结果:腹腔葡萄糖耐量和丙酮酸耐量试验结果显示,与正常组比较,模型组各时间点的血糖和曲线下面积(AUC)显著增加(P0.01);与模型组比较,健脾清化方能降低各时间点的血糖和AUC(P0.05)。肝糖异生相关蛋白及mRNA结果显示,与正常组比较,模型组大鼠肝脏pFOXO1/FOXO1蛋白表达的比值显著降低(P0.01),G6Pase、PEPCK蛋白及mRNA表达显著增加(P0.01);与模型组比较,健脾清化方组大鼠肝脏pFOXO1/FOXO1蛋白表达的比值显著增加(P0.01),G6Pase、PEPCK蛋白及mRNA表达降低(P0.05)。结论:2型糖尿病大鼠肝脏糖异生增加;健脾清化方可通过影响2型糖尿病大鼠肝脏糖异生相关酶的表达,减少其肝脏糖异生,从而降低血糖。
