摘 要: | 目的 :探讨乳腺癌组织中基质金属蛋白酶 2 ( MMP- 2 )、基质金属蛋白酶 9( MMP- 9)及金属蛋白酶 1组织抑制剂 ( TIMP- 1 )、金属蛋白酶 2组织抑制剂 ( TIMP- 2 )的表达及它们与乳腺癌生物学行为的关系。方法 :应用免疫组织化学 SP法 ,检测 49例乳腺癌、1 0例癌旁正常组织中MMP- 2、MMP- 9、TIMP- 1及 TIMP- 2的表达及分布。结果 :乳腺癌组织中 MMP- 2、MMP- 9、TIMP- 1及 TIMP- 2的表达明显高于癌旁正常组织 ,二者之间差异具有显著性 ( P<0 .0 1 ) ;乳腺癌组织中 MMP- 2、MMP- 9及 TIMP- 1主要在肿瘤细胞胞质内表达 ,而 TIMP- 2表达于瘤细胞和间质细胞胞质 ;乳腺癌组织中 MMP- 2、MMP- 9的表达与组织学分级、淋巴结转移密切相关( P<0 .0 5 ) ,能促进乳腺癌的侵袭和转移 ,而 TIMP- 1、TIMP- 2的表达与肿瘤生物学行为无相关性( P>0 .0 5 )。结论 :基质金属蛋白酶可成为判定乳腺癌生物学行为和指导临床治疗的可靠指标

Significance of Expression of Matrix Metalloproteinases and Their Inhibitors in Breast Carcinoma |

Abstract: | Objective: To study on the relationship between the expression of matrix metalloproteinase 2 (MMP 2) ,matrix metalloproteinase 9(MMP 9) and tissue inhibitor of metalloproteinases (TIMP 1 and TIMP 2) and biological behaviors in breast carcinoma. Methods:The expression and distribution of MMP 2, MMP 9,TIMP 1 and TIMP 2 were detected in 49 cases of breast carcinoma and 10 cases of corresponding non tumor normal tissues by immunohistochemical technique as a control. Results:In comparison with corresponding non tumor normal tissue group, the expression of MMP 2,MMP 9, TIMP 1 and TIMP 2 in breast carcinoma group was significantly higher ( P <0.01). MMP 2, MMP 9, and TIMP 1 were expressed predominantly in tumor cytoplasm, but positive staining of TIMP 2 was seen in the tumor and stroma cytoplasm. The expressions of MMP 2 and MMP 9 were correlated with histological grade and lymph node metastasis ( P <0.05), MMPs may promote tumor invasion and metastasis. There was no relationship between the expression of TIMP 1 or TIMP 2 and tumor biological behavior. Conclusion:MMPs may be a reliable indicator to determine biological behaviors and clinical therapy of breast carcinoma. |
