针刺和中药对乳腺增生的抑制作用研究

摘    要:目的]研究针灸和中药对乳腺增生的抑制作用。方法]将实验大鼠随机分为正常组、模型组、针剌组、中药组、针刺加中药组(简称针药组)以及三苯氧胺西药组(简称西药组)共6组;除正常组外,以肌注苯甲酸雌二醇和黄体酮造成乳腺增生大鼠模型;对除模型组外的4个治疗组分别施以针刺、中药、中药加针剌和三苯氧胺的治疗措施;检测各组治疗前后的乳头高度,计算治疗后大鼠第2对乳腺标本在光镜下的乳腺每小叶腺泡数、腺泡腔直径及导管直径情况,并采用免疫组化法检测各组乳腺组织雌激素受体(ER)及孕激素受体(PR)的表达情况。结果]模型组乳头高度比正常组高,乳腺导管扩张,小叶、腺泡数明显增加,乳腺组织ER受体和PR受体表达的计分值也比正常组高(均P<0.01),说明造模成功;经治疗,针刺、中药、针刺加中药以及三苯氧胺均使乳头高度降低(均P<0.01),乳腺每小叶腺泡数、腺泡腔直径及导管直径减少(P<0.05或P<0.01),乳腺组织ER、PR受体表达降低(P<0.05或P<0.01),但均未恢复到正常组水平(均P<0.05);其中针药组和西药组对乳腺增生的抑制作用均比单纯针刺组或单纯中药组略优,针药组和西药组两者作用相仿。结论]过度雌激素的刺激可能主要通过ER、PR的介导使大鼠乳腺组织增生,针刺、中药和西药三苯氧胺均对乳腺增生有抑制作用,而针

Inhibitory Effect of Acupuncture Combined with Herbal Medicine on Hyperplasia of Mammary Glands in Rats

Abstract:Objective] To observe the inhibitory effect of acupuncture combined with herbal medicine on hyperplasia of mammary glands in rats. Methods] Rats were randomized into 6 groups: A (normal) , B (model) , C (acupuncture) , D ( herbal medicine), E (acupuncture combined with herbal medicine) and F (triphenyloxamine) . Rat models of hyperplasia of mammary glands in Group B, C, D, E and F were established by intramuscular injection of estradial benzoate and progesterone. Nipple height in rats were detected before and after treatment. The number of acini in each lobule, the diameter of acinar cavity and duct in the second pair of mammary glands were calculated under light microscope. The expression of estrogen receptor (ER) and progesterone receptor (PR) in the tissues of mammary gland were examined by immunohistochemical method after treatment. Results] Nipple height in group B was more than that in group A and the scores of ER and PR expressions were also higher (P < 0.01), indicating the accomplishment of modeling. After treatment, nipple height was lessened ( P < 0.01) ; number of acini, diameter of acinar cavity and duct were reduced (P<0.05 or P < 0.01) and ER and PR expressions were decreased in group C, D, E and F ( P < 0.05 or P < 0.01), while the above indexes still differed from those in group A (P < 0.05). The inhibitory effects of acupuncture combined with herbal medicine and triphenyloxamine were stronger than that of acupuncture and herbal medicine alone, the effect of the combination was similar to triphenyloxamine. Conclusion] Excessive hormone level can stimulate the hyperplasia of mammary glands in rats by the mediation of ER and PR; acupuncture, herbal medicine and triphenyloxamine can inhibit the hyperplasia of mammary glands; the effect of combination of acupuncture and herbal medicine is stronger than that of alone and is similar to triphenyl oxamine but without adverse effects.
