葡萄籽提取物抗动脉粥样硬化形成的实验研究

摘    要:目的:探讨葡萄籽提取物对C57BL/6J小鼠动脉粥样硬化形成的影响。方法:将48只C57BL/6J小鼠,随机分为正常对照组(饲普通饲料)、高胆模型组(饲高脂饲料:含2%胆固醇、0.4%胆酸盐、10%猪油)、葡萄籽提取物低,高剂量组(在饲高脂饲料的同时,分别添加葡萄籽提取物0.2,0.6mg/g体重)。实验至第21周末,眼球放血处死小鼠,检测血清甘油三酯(TG)、胆固醇(Chol)、氧化型低密度脂蛋白(OX-LDL)和红细胞丙二醛(MDA)、超氧化物歧化酶(SOD)活性,同时取主动脉作病理形态学观察。结果:与高脂模型组比较,葡萄籽提取物组小鼠血清TG、Chol、OX-LDL、MDA含量明显降低,而红细胞SOD活性明显提高(P<0.01)。同时葡萄籽提取物组小鼠主动脉壁厚度和泡沫细胞数量明显较高脂模型组减轻,小鼠主动脉损伤评分差异显著P<0.01。结论:葡萄籽提取物具有减缓C57BL/6J小鼠动脉粥样硬化形成的作用,其作用可能与降低TG、Chol、OX-LDL水平,抗脂质过氧化作用有关。

Abstract:Objective:To observe the anti-atherosclerosis and an tioxidation effect of the extract of grape stones.Methods:48C57/6J mice were di vided randomly into4groups:the control group,the hy-perlipi demia mod el,the growpgiven low dosage of grape stone extract and the groupgiven high dosage.Serum triglycerides(TG),cholesterol(Chol),su peroxide dis mu tase(SOD),malonyldialdehyde(MDA),Oxidized low den sity lipopro tein(OX-LDL)were mea sured in the21rst week,and the form of aortic valves were ob-served pathologically Re sults:Com pared with those in the hyperlipi demia mod el group,the levels of MDA and OX-LDL in the serum of mice given grape stone extract were lower,while the activ ity of SOD in RBC of mi ce given grape stone ex tract was in creased (P<0.01).The thick ness of aortic valves in mice fed with grape stone extract was thinner than and the number of foam cells lower,in the hyperlipi demia model.Con clu sion:The extract of grape stone has in hi bi tory effect against atherosclerosis in C57BL/6Jmice,the possible mechanism may be related to the decrease of TG,Chol,OX-LDL and anti-peroxidation.
