食管鳞癌及不典型增生组织中端粒酶活性的定性及定量检测

摘    要:目的 :探讨食管鳞癌及不典型增生组织中端粒酶活性检测的意义及其与临床病理因素的关系。方法 :采用TRAP -ELISA定量及TRAP -银染定性的方法 ,对 38例食管鳞癌组织、15例不典型增生组织及 12例正常食管粘膜组织进行端粒酶活性检测。结果 :食管鳞癌、不典型增生组织及正常食管粘膜的端粒酶阳性表达率分别 84 .2 1%(32 / 38)、6 0 .0 0 % (9/ 15 )及 8.33% (1/ 12 )。端粒酶活性的平均A值分别为食管鳞癌组织 1.4 4 0± 0 .85 7、不典型增生组织 0 .4 86± 0 .30 2、正常食管粘膜组织 0 .0 75± 0 .0 4 9。 3组间两两比较端粒酶阳性表达率及端粒酶活性的平均A值均存在显著性统计学差异。结论 :端粒酶参与了食管癌的发生与发展 ,端粒酶的激活是食管癌的早期事件 ,其活性的检测有望成为预测食管癌发生及其诊断的良好指标。

Abstract:Objective: To investigate the relationship between telomerase activity and clinical pathologic factors of human esophageal squamous cell carcinomas(ESCC) and atypical hyperplasia tissues of the esophagus. Methods: Telomerase activity in specimens from 38 patients with ESCCs,15 patients with atypical hyperplasia tissue and 12 patients of normal esophageal mucose was measured quantitatively by TRAP-ELISA and qualitatively by TRAP-silver staining. Results: It was found that telomerase activity was positive in 32 of the 38 ESCCs specimen (84.21%),in 9 of the 15 atypical hyperplasia tissue specimen (60.00%) and in 1 of the 12 normal esophageal mucose(8.33%).The A value of telomerase activity in ESCC, atypical hyperplasia tissue and normal esophageal mucose were 1.440±0.801, 1.090±0.631 and 0.376±0.211 respectively. There was statistically significant difference between ESCC , atypical hyperplasia tissue and normal esophageal mucose in teolmerase positive experession and the average A value of talomerase. Conclusion: Telomerase is involved in carcinogenesis and malignant progression of ESCC. Activation of telomerase is the early event in ESCC, and telomerase activity might be a good marker in estimating the occurrence, development and diagnosis of ESCC.
