摘    要:目的 研究5-氟尿嘧啶(5-fluorouracil,5-FU)体外诱导胃癌SGC-7901细胞凋亡作用,及细胞凋亡与Ki-67抗原、核增殖抗原(PCNA)、P53蛋白、Bax蛋白、死亡受体Fas表达的关系。探讨5-FU诱导胃癌细胞凋亡作用的机制。方法 应用流式细胞仪和3’-OH末端标记法(TUNEL)检测凋亡细胞,电镜观察凋亡细胞的形态改变,免疫组织化学法检测细胞Ki-67、PCNA、P53、B

Abstract:Objective To study the effect of 5 FU which induced apoptosis of the gastric carcinoma cell line SGC 7901 in vitro and the relationship between apoptosis induced by 5 FU and the expressions of Ki 67, PCNA,P53,Bax,Fas of cells, and to investigate the mechanism of apoptosis of gastric carcinoma cell line SGC 7901 induced by 5 FU.Methods The apoptosis index(DI)of cells was evaluated by in situ TUNEL and Flow Cytometry and the apoptotic cells were studied by electron microscopy morphologically. The expressions of Ki 67,PCNA,P53,Bax and Fas of cells were detected by the method of immunohistochemistry.Results The DI of the experimental groups was significantly enhanced, which was associated with time and dosage and with the expression of apoptosis related gene Bax. The expressions of Ki 67, PCNA of cells of experimental groups were lower than those of control group. The expressions of P53 and Fas were not found in both experimental and control groups.Conclusion This study suggests that the apoptosis may be induced in gastric carcinoma cell line SGC 7901 by 5 FU and that the apoptosis be independent pathway of the P53.
