心康冲剂对慢性心衰大鼠AQP2mRNA、AVPV2mRNA表达的影响

摘    要:目的 观察心康冲剂对慢性心衰大鼠AQP2mRNA、AVPV2mRNA表达的影响,探讨其对慢性心衰水潴留的作用机制.方法 70只SD大鼠随机抽取10只为正常对照组,其余大鼠腹腔注射阿霉素(2 mg/kg,1次/周,共10周)造成心衰模型,再随机分为模型对照组,芪苈强心组,心康冲剂低、中、高剂量组.药物干预6周后,各组随机取6只存活大鼠,检测肾脏AQP2mRNA、AVPV2mRNA表达水平的变化.结果 与正常组相比,模型组AQP2mRNA、AVPV2mRNA的表达上升,差异有统计学意义;与模型组相比,芪苈强心组、心康冲剂各组均可显著下调AQP2mRNA、AVPV2mRNA的表达(P<0.05);与芪苈强心组相比,心康冲剂各组下调AQP2mRNA、AVPV2mRNA表达更为明显,差异有统计学意义(P<0.05).心康冲剂各组间比较无显著性差异(P>0.05).结论 心康冲剂可下调慢性心衰大鼠AQP2mRNA、AVPV2mRNA的表达,可能是其利尿消肿的作用机制之一.

Abstract:Objective The effect of Xinkang granule on AQP2mRNA and AVPV2mRNA in chronic heart failure rats was observed. This paper also discussed the mechanism of its water retention in chronic heart failure. Methods 70 SD rats were randomly assigned 10 rats as normal control group, the other rats by intraperitoneal injection of doxorubicin (2 mg/kg, 1 time/week for 10 weeks) caused by heart failure model were randomly divided into model group,Qiliqiangxin group, Xinkang granule low, medium and high dose groups. After six weeks drug intervention, six surviving rats randomly taken from each group were detected the expression level of AQP2mRNA and AVPV2mRNA in kidney. Results Compared with the normal group, the expressions of AQP2mRNA and AVPV2mRNA in model group increased a statistically significant difference. Compared with model group, the expressions were significant down regulated in Qiliqiangxin group and Xinkang granule groups (P〈0.05). Compared with Qiliqiangxin group, each group with Xinkang granule showed a more down-regulated AQP2mRNA and AVPV2mRNA expressions; there were significant differences (P〈0.05). There was no remarkable differences among each Xinkang granule groups (P〉0.05). Conclusion Xinkang granule can down-regulate the expressions of AQP2mRNA and AVPV2mRNA inchronic heart failure rats that may be one of the mechanisms of its diuretic swelling.
