毛囊与体表来源上皮细胞的生物学差异

摘    要:目的:观察毛囊及表皮来源上皮细胞之间的生物学差异,探讨其在创面愈合中的不同作用。方法:体外分离培养成人头皮毛囊及肢体皮肤来源的上皮细胞,通过β1-整合素及keratin15染色鉴定、细胞生长曲线测定、细胞克隆形成率比较及流式细胞仪分析等方法,比较两种细胞在细胞表面标志物、生长增殖、细胞周期等方面的生物学差异。结果:毛囊来源的上皮细胞的β1-整合素及keratin15染色均可见明显的阳性细胞,而体表皮肤来源的上皮细胞则多为阴性;在细胞的生长增殖方面两者存在一定的差异,但无统计学意义;细胞周期分析表明,毛囊来源的上皮细胞处于DNA合成期的细胞数明显高于体表皮肤来源的上皮细胞(23.44%傩11.33%,P〈0.05);毛囊来源的上皮细胞的克隆形成率为15.3%,后者为6.7%,差异显著。结论:毛囊来源的上皮细胞在增殖潜力方面明显高于体表皮肤来源的上皮细胞,在创面愈合中可能起主导作用。

Abstract:AIM: To investigate the different characteristics between the keratinocytes derived from follicles and those from epidermis and to discuss their effect in the wound healing process. METHODS: The keratinocytes were harvested respectively from the scalp follicles and the skin epidermis. Having been cultured in K-SFM serum free culture medium in vitro, the keratinocytes were identified by β1-intergrin and keratin15. The third passage was used to explore the characteristics of the growth kinetic. The cell cycle was determined with flow cytometry and the colony forming efficiency (CFE) was studied. RESULTS: The keratinocytes derived from the follicles were positive by β1-intergrin and keratin15 staining while the keratinocytes from the epidermis were mostly negative. The cell cycle profile analysis showed that the ratio of S stage in the keratinocytes from the follicles was significantly higher than that in the keratinocytes from the epidermis (23. 44% vs 11.33%, P〈0.05). The CFE of the keratinocytes from the follicles was 15.3% and that of the control group was 6.7% ( P 〈 0.05 ). CONCLUSION : The keratinocytes derived from, the follicles have some characteristics of the epidermal stem cells and their proliferative potential is higher compared with that of the keratinocytes from the skin epidermis.
