摘    要:目的:研究格列卫和六亚甲基二乙酰胺(HMBA)对K562细胞增殖、凋亡、细胞周期及周期蛋白表达的影响.方法:MTT法观察细胞生长指数和细胞存活率,流式细胞仪、电镜检测细胞周期和凋亡,半定量RT-PCR检测bcr-abl基因的表达,Western blotting检测蛋白表达.结果:联合应用两种药物后细胞存活率明显下降,凋亡比例略下降,细胞周期阻滞于G0/G1期,cyclinD1、cyclinE、cyclinA、CDK4、c-myc蛋白表达下调.结论:两种药物应用后出现细胞存活率下降、细胞周期阻滞、周期蛋白表达下调、凋亡比例下降,联合作用对细胞的增殖抑制具有协同作用,进入凋亡途径细胞减少。

Effects of Glivec and hexamethylenebisacetamide on proliferation and apoptosis of K562 cells
Abstract:AIM: To explore how Glivec and HMBA affect the cell growth, apoptosis, cell cycle and cyclin protein expression of K562 cells. METHODS: After K562 cell line was cultured and Glivec and/or HMBA were added, the cell growth index and surviving rate were determined with MTT assay, the cell cycle and apoptosis were examined by flow cytometry and electron microscopy, the expression of bcr abl gene was examined by RT PCR, and the expressions of cyclinD1, cyclinE, cyclinA, CDK4 and c myc protein were examined by Western blot. RESULTS: A combination of two drugs apparently reduced the surviving rate and slightly declined the percent of apoptosis. K562 cells were arrested in G0/G1 phase, and the expressions of cyclinD1, cyclinE, cyclinA, CDK4 and c myc protein were downregulated. CONCLUSION: A combination of two drugs can effectively inhibit the growth and slightly inhibit the apoptosis of K562 cells. The action of drugs bears on cell cycle arrest, and downregulation of cyclin protein and cell apoptosis.
