微滤-大孔树脂法与醇沉法精制苦参中氧化苦参碱和苦参总黄酮的比较研究

摘    要:目的:考察微滤-大孔树脂法精制苦参中氧化苦参碱、苦参总黄酮的效果,并与水提醇沉法进行了比较。方法:采用HPLC法测定氧化苦参碱的含量,紫外分光光度法测定苦参总黄酮的含量。结果:苦参提取液经微滤-大孔树脂法处理后,其氧化苦参碱、苦参总黄酮的保留率为78.88%和73.4%,固形物去除率为38.95%;经醇沉法处理后,氧化苦参碱、苦参总黄酮的保留率为60.7%和52.1%,固形物去除率为34.88%。结论:微滤-大孔树脂法较醇沉法可更有效地保留有效成分、去除杂质。

Abstract:Objective:To study the effect of microfiltration macroporous resin method in refining of oxymatrine and general flavone in sophora flavescens,and to compare the results with that of alcohol sedimentation. Methods: HPLC was applied to analyze the content of oxymatrine in the samples. UV spectrophotometry was applied to analyze the content of general flavone. Results:In sophora flavescens decoction,the recovery rates of oxymatrine and general flavone were 78.88% and 73.4% respectively and the whole solid was decreased by 38.95% by microfiltration macroporous resin method; the recovery rates of oxymatrine and general flavone were 60.7% and 52.1% respectively and the whole solid was decreased by 34.88% by alcohol sedimentation method. Conclusion:Microfiltration macroporous resin method was more effective than alcohol sedimentation for refining oxymatrine and general flavone in sophora flavescens.
