G-CSF对血管性痴呆大鼠海马Ach含量及CHAT活性的影响

摘    要:目的探讨粒细胞集落刺激因子(G-CSF)干预对血管性痴呆(VD)大鼠的作用及机制。方法 54只SD大鼠随机分为3组:假手术组、VD模型和G-CCF组各18只,后2组采用永久性双侧颈总动脉结扎法建立大鼠VD模型,G-CSF组大鼠每日皮下注射G-CSF 50μg/kg,术后7、14、28 d用Morris水迷宫进行定向航行观察大鼠逃避潜伏期,评价大鼠空间学习记忆能力,Western Blot检测大鼠海马区脑组织Ach、CHAT蛋白表达的变化。结果 (1)逃避潜伏期变化:术后7、14、28 d,与假手术组比较,VD模型组和G-CSF组平均逃避潜伏期均延长(P<0.01);与VD模型组比较,G-CSF组平均逃避潜伏期缩短(P<0.01)。(2)Ach、CHAT蛋白表达:术后7、14、28 d,与假手术组比较,VD模型组Ach、CHAT蛋白表达均明显减少(P<0.01);与VD模型组比较,G-CSF组Ach、CHAT蛋白表达均明显升高(P<0.01)。结论 G-CSF促进血管性痴呆大鼠海马区Ach、CHAT蛋白表达,改善大鼠学习记忆功能。

Abstract:Objective To investigate the mechanism of granuloctye colony stimulating factor(G-CSF) in the intervention of vascular dementia(VD) rats.Methods Fifty-four aged rats were randomly divided into sham group,VD model group and G-CSF intervention group.The VD model group and G-CSF group was set up with a permanent bilateral occlusion of both common carotid arteries for establishing vascular dementia models.The G-CSF intervention group rats were given G-CSF 50μg/kg every day after operation by hypodermic injection.Morris water maze test was conducted to observe the rats escape latency and evaluate the spatial learning and memory abilities.Western Blot was applied to detect the expression of Ach, CHAT in rat hippocampus.Results(1) The change of escape latency:compared with the sham group,the average duration of escape latency was prolonged in the groups of the VD model and G-CSF intervention(P<0.01 ).Compred with the VD model group,the average duration was shorten afger G-CSF internvented(P<0.01 ).(2 ) The expression of Ach,CHAT; compared with the sham group,the expression of Ach,CHAT in the hippocampus of the VD model group and the G-CSF intervention group was significantly increased at the time points of 7,14,28 days(P<0.01 ).Compared with the VD model group,the expression levels were increased in the group of G-CSF interventional(P<0.01).Conclusion G-CSF could promoted the expression of the Ach,CHAT in the hippocampus,and improve the learning and memory ability in vascular dementia rat.
