维生素D联合葛根素对四氯化碳致大鼠肝纤维化的作用及机制

摘    要:目的 探究维生素D联合葛根素对肝纤维化的保护作用及机制.方法 将大鼠分为正常对照组(C组)、四氯化碳组(CCl4组)及CCl4外加维生素D组(V组)、葛根素组(P组)及维生素D联合葛根素组(V+P组)治疗组,8周后处死,检测透明质酸(HA);测量羟脯氨酸(Hyp);制作肝脏石蜡切片并行天狼星红染色检查;反转录PCR检测肝组织中胶原蛋白(Collagen)I、ⅢmRNA水平;Western blot检测肝脏NF-κB和肿瘤坏死因子(TNF-α)水平.结果 CCl4组出现明显肝脏纤维化,V组、P组及V+P组均明显改善了纤维化程度,降低了血中HA水平及肝脏中的Hyp水平,肝脏Collagen Ⅰ、ⅢmRNA及NF-κB和TNF-α蛋白水平明显降低.其中V+P组肝纤维化程度改善最为明显(P<0.05).结论 维生素D联合葛根素可以保护CCl4引起的大鼠肝脏纤维化,其机制可能与降低肝脏星状细胞活化、减少胶原纤维分泌有关.

Abstract:Objective To explore the protective effect and mechanism of vitamin D combined with puerarin on liver fibrosis.Methods The rats were divided into normal control group (C),tetrachloromethone group (CCl4),vitamin D group (V),puerarin group(P) and vitamin D combined with puerarin group(V+P).After 8 weeks,the rats were sacrificed and blood and liver samples were collected.The level of blood hyaluronic acid(HA) was tested.The hydroxyproline(Hyp) level in the liver was measured.The liver paraffin sections were made and examined by the sirius red staining.The mRNA levels of collagen Ⅰ and collagen Ⅲ in the liver tissue were detected by RT-PCR,and the levels of NF-κB and TNF-α in the liver were detected by Western blot.Results The CCl4 group appeared obvious liver fibrosis.The liver fibrosis degree was significantly improved in the group V,P and V+P,the blood HA level and liver Hyp level were reduced.The mRNA levels of collagen Ⅰ and collagen Ⅲ as well as the protein levels of NF-κB and TNF-α in the liver were significantly decreased.Among them.The liver fibrosis improvement degree in the V+P group was most significant.Conclusion Vitamin D combined with puerarin can protect rat liver fibrosis induced by CCl4 and its mechanism may be related with reducing the activation of hepatic stellate cells(HSC) and decreasing the collagenous fibers secretion.
