积雪草甙、羟基积雪草甙对体外培养人胚胎成纤维细胞的影响

摘    要:目的研究积雪草甙(asiaticoside,Ad)和羟基积雪草甙(madecassoside,Md)对体外培养人胚胎成纤维细胞(Human embryonic skin fibroblasts,HESFb)增殖过程的影响。方法在不同浓度(0μg/ml、10μg/ml、50μg/ml、100μg/ml、200μg/ml、400μg/ml)Ad和Md的环境下对人胚胎细胞进行体外培养,用CCK-8试剂盒通过酶标仪检测不同培养时间(0、1、2、3、4、5d)HESFb的OD值,用SPSS13.0统计软件进行统计分析。结果在实验时间内Ad和Md在10μg/ml、50μg/ml、100μg/ml、200μg/ml浓度时均对人胚胎成纤维细胞的增殖有促进作用,200μg/ml浓度时最明显,且Ad比Md作用强。两组在400μg/ml浓度时均有抑制作用。结论在一定浓度和培养时间内一定剂量的Ad和Md对体外培养的HESFb增殖有促进作用,同剂量相比Ad比Md明显。而较高剂量的时候均可出现抑制作用。

Abstract:Objective To investigate the effects of asiaticoside(Ad) and madecassoside(Md) on the proliferation of human embryonic skin fibroblasts(HESFB) in vitro.Methods Culture HESFB under different concentration condition(0μg/ml,10μg/ml,50μg/ml,100μg/ml,200μg/ml,400μg/ml) of Ad and Md in virto,detect the OD value of HESFB in different incubation time(0 day,2 days,3 days,4 days,5 days) by Cell Counting Kit-8(CCK-8) through ELIASA,and the data was analyzed with SPSS13.0 statistic software.Results Ad and Md can stimulate proliferation of HESFB when at 10μg/ml,50μg/ml,100μg/ml and 200μg/ml within 5 days,especially at 200μg/ml,and Ad is more effective than Md.Both of them appeared inhibitory effects at 400μg/ml.Conclusion In a certain concentration range and time,Ad and Md can stimulate proliferation of HESFB in vitro,and the effect of Ad is better than Md under the same condition.While inhibitory effects appeared under higher concentration condition.
