| 摘 要： | 目的:探讨皮质酮致大鼠胸腺细胞功能退化可能的机制及左归丸的药理作用。方法:大鼠胸腺细胞体外培养,分正常对照组、模型组、左归组。MTT法检测细胞增殖情况,PI染色结合FCM分析周期分布,Real time PCR和Western blotting法检测相关因子表达变化。结果:皮质酮显著抑制细胞增殖,细胞阻滞于G1期,p16表达显著升高(P0.01),cdk4、IL-2、IL-2Rα蛋白表达降低(P0.05)。与模型组比较,左归组G1期细胞数减少,p16表达降低,IL-2、IL-2Rα蛋白表达升高(P0.05)。结论:左归丸通过抑制p16表达,促进细胞增殖,延缓皮质酮致胸腺依赖性免疫功能退化的进程。
