活血化瘀中药干预兔动脉粥样硬化形成作用研究

摘    要:目的观察活血化瘀中药丹参、川芎、姜黄对兔动脉粥样硬化(AS)的影响及其作用机制。方法将家兔按血脂水平随机分为空白组、模型组、阿托伐他汀组、丹参组、川芎组及姜黄组。其中空白组给予普通饲料,其余各组饲喂高脂饲料。高脂饲料喂养期间各组动物分别给予相应剂量的药物。给药第12周后,测定血脂水平、血清抗氧化能力、凝血及血液流变学指标,并观察动脉斑块形成程度。结果川芎与丹参可显著降低AS兔主动脉弓部斑块校正面积与最大斑块厚度。川芎、丹参、姜黄均可降低AS兔血小板最大聚集率,丹参可显著降低AS兔低切全血黏度,川芎可降低AS兔血清总胆固醇、低密度脂蛋白胆固醇及非高密度脂蛋白胆固醇水平。川芎、丹参可降低血清脂质过氧化产物丙二醛水平,丹参还可升高超氧化物歧化酶的活性。结论活血化瘀中药具有较好的抑制AS形成的作用,与其降血脂、提高抗氧化能力等作用有关。

Abstract:Objective To observe the effect and mechanism of Radix et Rhizome Salviae Miltiorrhizae,Rhizoma Chuanxiong and Rhizoma Curcumae Longae,which possessing activating blood circulation to dissipate blood stasis function,on atherosclerosis of rabbits.Methods The rabbits were randomly divided into six groups:blank control group,model control group,atorvastatin group,Radix et Rhizome Salviae Miltiorrhizae group,Rhizoma Chuanxiong group and Rhizoma Curcumae Longae group,according to the serum lipid level.The rabbits in blank control group took ordinary feeds and the others tooks high-fat feeds.The rabbits were given corresponding dose of the drug for twelve weeks.Then the serum lipid level,serum antioxidant ability,blood coagulation,hemorheology were measured,and the pathological arteriosclerosis plaque lesions were observed.Results Radix et Rhizome Salviae Miltiorrhizae and Rhizoma Chuanxiong reduced the plaque area corrected by cross-sectiona vessel wall area(PA/CVA) and the max plaque thickness(MPT) at the aortic arch in arteriosclerosis rabbits.All of the three drugs decreased the max platelet aggregation rate(MPAR),and Radix et Rhizome Salviae Miltiorrhizae also reduced low shear blood viscosity.Rhizoma Chuanxiong reduced the serum cholesterol(CHO),low-density lipoprotein(LDL-C) and non-high density lipoprotein(non-HDL-C) level in arteriosclerosis rabbits.Radix et Rhizome Salviae Miltiorrhizae and Rhizoma Chuanxiong reduced the serum malondialdehyde(MDA) level,and Radix et Rhizome Salviae Miltiorrhizae also increased the serum superoxide dismutase(SOD) activity.Conclusion The traditional Chinese medicines which possessing activating blood circulation to dissipate blood stasis function can inhibit the atherosclerosis formation by reducing blood lipids,promoting anti-oxidant ability.
