茉莉酸甲酯对丹参毛状根中水溶性酚酸类化合物积累的影响

摘    要: 目的 研究茉莉酸甲酯(methyl jasmonate,MJ)对丹参毛状根生长和其中丹参素、迷迭香酸、丹酚酸B 3种化合物含量的影响。方法 以MJ浓度和添加时间为2个影响因素,设计5个水平的正交实验,观察不同MJ浓度和添加时间对丹参毛状根生长和丹参素、迷迭香酸、丹酚酸B 3种化合物含量的影响。结果 MJ的加入对丹参毛状根生长有一定抑制作用,并能显著改变丹参素、迷迭香酸、丹酚酸B的含量。在接种第15天添加浓度为300 μmol·L-1MJ、共培养15 d的毛状根中丹参素含量达(0.510±0.054)%,约为对照样品(0.179±0.02)%的2.85倍;接种第20天加入浓度为300 μmol·L-1MJ、共培养10 d的毛状根中迷迭香酸的含量达(14.358±0.710)%,约为对照样品(7.021±0.403)%的2倍;接种0天时添加200 μmol·L-1 MJ、共培养30 d的丹参毛状根中丹酚酸B的含量高达(1.592±0.120)%,约为对照样品(0.189±0.05)%的8.42倍。结论 茉莉酸甲酯能明显改变丹参素、迷迭香酸、丹酚酸B在丹参毛状根中的积累,并且随MJ浓度和添加时间的改变呈现一定规律性,这对丹酚酸类化合物在植物体内生物合成途经的研究有一定借鉴意义。

Abstract:OBJECTIVE To study the effects of methyl jasmonate (MJ) on the accumulation of the main water-soluble phenolic acids in Salvia miltiorrhiza hairy roots. METHODS The different dosages of MJ were added into the suspension cultures at different growth stages of S. miltiorrhiza hairy roots. The contents of Danshensu, rosmarinic acid,and salvianolic acid B were determined by HPLC. RESULTS MJ affected the accumulation of these compounds in a dose-dependent manner. The MJ adding time also showed significant influence on the accumulation. The peak content of Danshensu was (0.510±0.054)% by 300 μmol·L-1 MJ cultured with the hariy root for 15 d. The peak contents of rosmarinic acid was (14.358±0.710)% by adding 200 μmol·L-1 MJ culture with the hariy root for 10 d. The peak content of salvianolic acid B was (1.592±0.120)% by 200 μmol·L-1 MJ cultured with the hariy root at the beginning. CONCLUSION MJ could obviously affect the accumulation of Danshensu, rosmarinic acid, salvianolic acid B,as well as the growth of the hairy roots. It is helpful for the further research of the biosynthesis of phenolic acids in Salvia miltiorrhiza Bunge.
