冠心Ⅱ号方含药血清对心肌细胞缺血再灌注样损伤的比较药效学研究

摘    要:目的:比较汤剂、微米制剂、有效成分重组复方三种冠心Ⅱ号方制剂的含药血清对体外培养心肌细胞缺血再灌注样损伤的影响,为临床使用和药物二次开发提供依据。方法:大鼠分别灌胃给予合生药剂量5g/kg的冠心Ⅱ号方汤剂、微米制剂或有效成分重组复方,1次/日连续5日,末次给药后90min采血制备含药血清。采用MTT法观察含药血清对体外培养大鼠正常心肌细胞活性及生长情况的影响,并检测其对缺氧-复氧模型或过氧化氢损伤心肌细胞培养上清乳酸脱氢酶(LDH)活性、肌酸激酶(CK)活性、丙二醛(MDA)含量、细胞胞浆超氧化物岐化酶(SOD)活性的影响。结果:三种冠心Ⅱ号方的含药血清对体外培养大鼠正常心肌细胞活性及生长无显著影响;而三者均能显著减轻由缺氧-复氧或过氧化氢造成的心肌细胞培养上清中LDH及MDA含量增高的变化,对细胞胞浆SOD活性降低有明显的保护作用,并可降低培养上清中CK活性,但三者比较未观察到显著性差异。结论:不同冠心Ⅱ号方的含药血清对体外培养心肌细胞缺血再灌注样损伤均有明显的保护作用,但三者作用差异不显著。

Abstract:Objective: To compare the effect of different officinal forms (such as medicinal broth, micrometer praeparatum and recombination of effective constitute) of Guanxin II formula on the ischemia-reperfusion injury in cultured rat myocardial cells to offer foundation for clinical apply and the further exploitation of the drug. Methods: The blood serum of drug was prepared at 90 min after the last administration when the five administrations of once a day at a dosage of 5g/kg. Myocardial cells of rat were cultured in vitro. The influence of the different blood serums in normal cultured rat myocardial cells and on the ischemia-reperfusion injury was observed. Resrlts: None of the blood serum of medicinal broth, micrometer praeparatum and recombination of effective constitute of Guanxin II formula did significant effect in normal cultured myocardial cells. And all subjects showed notable protection on the ischemia-reperfusion injury. The reduction of the activity of LDH and CK, as well as the decrease of the content of MDA was observed in the medium, and the damage of the ability of SOD in endochylema induced by ischemia-reperfusion injury was lessened. Though there was no apparent difference among them, the effect of recombination of effective constitute seemed better to some extent. Conclusion: The blood serum of different officinal form of Guanxin II formula showed no significant distinction in the protective effect on the ischemia-reperfusion injury in cultured rat myocardial cells.
