摘    要:目的观察非洛地平(Felo)和螺内酯(Spir)合用对高血压大鼠的动脉收缩压(SBP)、左室肥厚及左室心肌纤维化和血浆肾素、血管紧张素、醛固酮水平的作用。方法采用两肾一夹Goldblott高血压模型,尾容积法测定清醒大鼠动脉收缩压;分离左心室并称重,测定其羟脯氨酸,计算胶原蛋白含量。结果与模型对照组比较,Felo+Spir组SBP、左心室重与体重的比(LVW/W)及左心室胶原蛋白含量分别下降了44.5%,32.8%,32.0%;Felo组分别下降了36.6%,19.6%,10.3%。结论本实验结果提示,Felo和Spir在降低肾性高血压大鼠的SBP、抑制其左心室肥厚、心肌纤维化方面有协同作用。

Effects of felodipine and spironolactone combination in rat renovascular hypertension
Abstract:AIM To study the effects of felodipine and spironolactone combination on systolic blood pressure, left ventricular mass, myocardial fibrosis and plasma levels of renin, angiotensin Ⅱ and aldosterone in rat renovascular hypertension. METHODS Two kidney, one clip renovascular hypertension rats were randomly grouped. SBP was measured in conscious rats by tail cuff method. Cardiac content of collagen was assessed by determination of hydroxyproline. RESULTS In the felodipine treated group, SBP, LVW/W and left ventricular content of collagen were reduced by 366%, 196% and 103%, respectively, compared with the model group. In the spironolactone treated group, SBP and left ventricular content of collagen was lowered by 70% and 178%, respectively. In the combination treated group, SBP, LVW/W and left ventricular content of collagen were significantly lowered than either in the felodipine treated group or in the spironolactone treated goup. CONCLUSION The present results suggested that there was a synergism between felodipine and spironolactone in antagonizing left ventricular hypertrophy and myocardial fibrosis in rat renovascular hypertension.
