荞麦花叶总黄酮对大鼠急性压力负荷c-fos蛋白表达的影响

摘    要:目的:观察荞麦花叶提取的总黄酮(TFBFL)对大鼠急性压力负荷左心室c-fos蛋白表达的影响. 方法:通过腹主动脉狭窄建立心肌肥厚模型,术前给予TFBFL 0.1, 0.2和0.4 g/(kg/d),卡托普利 60 mg/(kg/d). 分别于狭窄后1,3和12 h处死动物取材. 用免疫组化方法检测左室心肌c-fos蛋白的表达. 结果:正常大鼠心肌没有或仅有少量c-fos表达,腹主动脉狭窄后1 h, c-fos表达明显增加,3 h达高峰,较对照组升高约3倍,12 h开始下降. 腹主动脉狭窄后3 h, TFBFL 0.4, 0.2和0.1 g/(kg/d)与心肌肥厚组相比, c-fos表达分别降低21.41%(P<0.01),14.52%(P<0.05)和10.36%. 结论:TFBFL可明显降低c-fos蛋白的表达.

Abstract:AIM: To explore the effect of total flavones of buckwheat flower and leaf (TFBFL) on the expression of c-fos protein in the course of acute pressure overload of the left ventricle in rats. METHODS: The rat models of myocardial hypertrophy caused by abdominal aortic banding were given ig TFBFL 0.1, 0.2 and 0.4 g/(kg/d)or captopril 60 mg/(kg/d) before operation. At 1, 3 and 12 h after operation,the expression of c-fos was detected by immunohistochemistry following the sacrification of all rats. RESULTS: The expression of c-fos in the left ventricular had a significant increase after 1 h of abdominal aortic banding and reached a peak at 3 h, decreased at 12 h. Compared with the models of myocardial hypertrophy, TFBFL 0.4, 0.2 and 0.1 g/(kg/d) reduced the expression of c-fos by 21.41% (P< 0.01), 14.52% (P<0.05) and 10.36% after 3 h of abdominal aortic banding. CONCLUSION: TFBFL can decrease the expression of c-fos in the course of acute pressure overload of the left ventricle in rats.
