康泰胶囊对肠易激综合征内脏高敏感性大鼠背根神经元兴奋性的影响

摘    要:【目的】观察康泰胶囊对肠易激综合征(IBS)内脏高敏感性大鼠结肠背根神经元(DRG)兴奋性的作用,探讨康泰胶囊治疗IBS的作用机制。【方法】选用SD大鼠,随机分为正常组、模型组、康泰胶囊组(剂量为6.06 g/kg)、匹维溴铵组(剂量为50 mg/kg),除正常组外,其他组大鼠均采用慢性不可预计条件应激法复制内脏高敏感性IBS模型,根据腹部回缩反射(AWR)结果评估模型内脏敏感程度,分离模型大鼠背根神经元,应用膜片钳全细胞记录技术测定背根神经元的基电流强度和2倍基电流刺激下神经元反应,观察康泰胶囊对内脏高敏感IBS大鼠结肠背根神经元电生理特性的影响。【结果】康泰胶囊可以显著降低模型大鼠AWR评分(P<0.05或P<0.01),显著升高结肠DRG的基强度(P<0.01),显著减少2倍基强度刺激产生的动作电位数(P<0.01)。【结论】抑制应激引起的内脏高敏感IBS大鼠结肠背根神经元兴奋性增高可能是康泰胶囊治疗IBS的作用机制之一。

Abstract:Objective To observe the effect of Kangtai Capsule on excitability of dorsal root ganglion(DRG) neurons of visceral hypersensitivity rats with irritable bowel syndrome(IBS),and to investigate the therapeutic mechanism of Kangtai Capsule for IBS.Methods We established the visceral hypersensitivity rat model through chronic unpredicted mild stress,and the visceral sensitivity to colorectal balloon distention(CRD) was measured by abdominal withdrawal reflex(AWR) scores.SD rats were randomized into normal group,model group,Kangtai Capsule group and western medicine group.Neurons were quickly dissociated from the dorsal root ganglion by a combination of mechanical and enzymatic method,and background current of colonic dorsal root ganglion as well as neuronal excitability stimulated by double background current was measured with whole cell patch clamp in rats before and after treatment with Kangtai Capsule.Results The AWR scores of rats in Kangtai Capsule group were much lower than those in the model group.After treatment with Kangtai Capsule,the background current was increased,and the incidence of action potential of colonic DRG neurons in model rats was reduced(P<0.01).Conclusion Kangtai Capsule has inhibitive effect on colonic DRG neuronal excitability of visceral hypersensitivity rats with IBS,which may be one of its therapeutic mechanisms for IBS.
