昆明种小鼠H2-Eb位点多态性与免疫性不孕不育的相关性研究

目的探讨昆明种小鼠H2-Eb位点多态性与免疫性不孕不育的关系。方法采用Freund精子完全佐剂与不完全佐剂免疫昆明种小鼠,复制免疫性不孕不育模型,ELISA法检测免疫小鼠抗精子抗体(AsAb),采用聚合酶链式反应序列特异性引物(PCR-SSP)技术扩增小鼠H2-Eb基因,琼脂糖凝胶电泳,SPSS 17.0统计软件分析AsAb(+)组与AsAb(-)组基因多态性差异。结果所有PCR-SSP的内参条带全部出现,AsAb(+)组、AsAb(-)组MudoEb5阳性检出率分别为65.79%(50/76)、67.74%(21/31),差异无统计学意义;AsAb(+)组、AsAb(-)组MudoEb7阳性检出率分别为30.26%(23/76)、61.29%(19/31),差异有统计学意义(P<0.05)。结论 MudoEb7可能是免疫性不孕不育昆明种小鼠的保护基因。

Correlation of KM mouse H2-Eb polymorphism and immune infertility

Objective To explore the correlation of H2-Eb polymorphism and immune infertility in KM mouse.Methods Freund compounds were applied to establish immune infertility model in KM mouse.ELISA was used to detected the level of anti-sperm antibody(AsAb).PCR-SSP method were used to amplify the second exon polymorphism of H2-Eb.Agarose gel electrophoresis and SPSS 17.0 software were adopted to analyze the difference between AsAb(+) group and AsAb(-) group.Results The internal positive control were all appeared.The detection rates of AsAb(+) group and AsAb(-) groups MudoEb5 positive were 65.79%(50 /76) and 67.74%(21 / 31) respectively,and no significant difference were found.While there was significant statistical difference on MudoEb7 positive between AsAb(+) group and AsAb(-) group with detection rates 30.26%(23 /76) and 61.29%(19/31) respectively(P < 0.05).Conclusion MudoEb7 allele may be a immune infertility protective gene in KM mouse.
