一种快速评价葛根药材质量的胶体金免疫层析试纸的研制

目的:研制快速评价葛根药材质量的胶体金试纸。方法:通过ELISA与HPLC检测结果的关联确定胶体金试纸的半定量限,通过对胶体金试纸制备过程的优化,确定最优胶体金制备方法。结果:葛根素胶体金试纸的半定量限为34μg/m L,胶体金试纸制备最佳工艺为胶体金标记最佳PH为8、抗体最佳标记量为0.48 mg/m L、胶体金溶液与抗体溶液最佳标记比例为2.5∶1。结论:葛根素胶体金试纸简便、快速、准确,可以满足对葛根药材的质量评价,尤其适用于原料采购现场的快速检测。

Development of a Colloidal Gold Immunochromatographic Test Paper for Rapid Evaluation of the Quality of Pueraria Lobata

Objective:To establish a colloidal gold test paper for rapid evaluation of pueraria lobata.Methods:To determine the semi-quantitative limit of the colloidal gold test paper through ELISA and HPLC,and to make sure the optimal colloidal gold preparation method by optimizing the process of colloidal gold test paper.Results:The semi-quantitative limit of the puerarin colloidal gold test paper is 34μg/mL,and the best preparation condition is that the best pH value for colloidal gold label is 8,the optimal value of antibody marker is 0.48mg/mL and the best markup ratio of colloidal gold solution and antibody solution is 2.5∶ 1.Conclusion:The puerarin colloidal gold test paper is simple,rapid and accurate,which can satisfy the quality evaluation of medical materials and be used for rapid detection of the raw material on the purchasing site.
