芸香苷的胃粘膜保护作用与NO、PG的关系

摘    要:目的 研究芸香苷(rutoside, Ru) 的胃粘膜保护作用与 N O 和 P G 的关系,方法 制备小鼠乙醇性胃粘膜损伤模型,测定胃粘膜损伤面积;测定小鼠胃组织中 N O、 P G E2 含量;评价 Ru 对 L N N A+ 体积分数为30 % 乙醇所致大鼠胃粘膜损伤的影响。结果  Ru 7 、14 、28 mg·kg - 1 ,ig ,bid ×5 d对无水乙醇诱发的小鼠胃粘膜损伤可产生剂量依赖性抑制作用,其损伤抑制率分别为206 % 、387 % 和522 % 。 Ru可使小鼠胃组织中降低的 N O 含量回升,但对 P G E2 含量无明显影响。 Ru 5 ,20 mg·kg - 1 ,ig ,亦可显著抑制由 N O S 抑制剂 L N N A+ 体积分数为30 % 乙醇所致的大鼠胃粘膜损伤。结论  Ru 对乙醇性胃粘膜损伤具有保护作用,其作用机制可能与促进胃粘膜细胞合成或释放内源性 N O 有关

Abstract:AIM To study the relationship between protective effect of rutoside (Ru) on gastric mucosa and nitric oxide (NO) and prostaglandin (PG). METHODS Make the model of gastric mucosal injury in mice induced by obsolute ethanol;the lesion area of gastric mucosa was measured; the contents of NO and PGE 2 of gastric tissue were determined;the effect of Ru on gastric mucosal injury induced by L NNA+30% ethanol was evaluated. RESULTS The lesion area of gastric mucosa in mice was inhibited dose dependently after administration of Ru (7, 14, 28 mg·kg -1 , ig, bid×5 d), and the inhibitory rates were 20 6%, 28 7% and 52 2% respectively .The decrease of NO content induced by ethanol was significantly elevated by Ru to normal level. but Ru did not obviously influence the content of PGE 2 of gastric tissue in mice. Ru (5,20 mg·kg -1 , ig, bid×5 d) could also significantly inhibit gastric mucosal injury in rats induced by NO synthase inhibitor L NNA+30% ethanol. CONCLUSIONS Ru and possesses the protective effect on gastric mucosal injury induced by ethanol and its mechanism may be related to improving synthesis and release of NO from gastric mucosal cells.
