金锁固精丸加味方对大鼠阿霉素肾病的防治作用

摘    要:【目的】观察金锁固精丸加味方对阿霉素肾病模型大鼠的治疗作用。【方法】将SD大鼠24只,随机分为正常对照组、模型组和金锁固精丸加味方组(中药组);除正常对照组外,其他组大鼠均采用一次性尾静脉注射阿霉素6 mg/kg复制肾病综合征模型;中药组同时给予金锁固精丸加味方水煎剂96g/kg灌胃,正常对照组、模型组以等容积饮用水灌胃,连续45 d;分别取24h尿、血清及肾组织观察该方对模型大鼠24h尿蛋白,血清总蛋白、白蛋白、总胆固醇、尿素氮、肌酐及肾组织形态学的影响。【结果】金锁固精丸加味方能降低24 h尿蛋白和血清总胆固醇,升高血清总蛋白和白蛋白,与模型组比较均有显著性差异(P<0.05或P<0.01),但对血清肌酐、尿素氮无明显影响(P>0.05);苏木素-伊红(HE)染色和电镜检查结果均显示该方可减轻模型大鼠肾组织病理改变。【结论】金锁固精丸加味方对阿霉素肾病模型大鼠具有一定的治疗作用。

Abstract:Objective] To observe the therapeutic effect of Modified Jinsuo Gujin Pill (MJGP) on adriamycin-induced nephropathy in rats. Methods] Twenty-four SD rats were randomized into normal control group, model group and MJGP group. Except the normal control group, rats in the other two groups were injected with one-dose adriamycin (6 mg/kg) into the caudal vein to induce nephrotic syndrome. Meanwhile, MJGP group was given MJGP decoction 96g/kg and the normal control group was given water of the same volume for 45 days. The effects of MJGP on the levels of 24-hour urine protein, serum total protein (TP), albumin (ALB), serum total cholesterol (TC), blood urea nitrogen (BUN) and serum creatinine (SCr), as well as on the renal histological features were observed. Results] MJGP decreased the levels of 24-hour urine protein and serum TC, and increased serum TP and ALB levels, the difference being significant as compared with the model group (P<0.05 or P<0.01). However, MJGP had no effect on SCr and BUN levels. HE stain results and examination under electron microscope showed that renal pathological changes were relieved in MJGP group. Conclusion] MJGP is effective for rat adriamycin-induced nephropathy in rats.
