骨肉瘤组织中survivin与VEGF的表达及相关关系的研究

目的检测survivin与VEGF在骨肉瘤中的表达,探讨二者对骨肉瘤血管生成和转移的作用及其关系.方法采用原位杂交、免疫组织化学技术检测38例骨肉瘤组织survivin mRNA表达及VEGF、CD34蛋白表达,计数微血管密度(MVD)、,并与主要临床病理参数进行比较.结果骨肉瘤中survivin mRNA和VEGF表达率均为65.8%,二者表达呈正相关(r=0.546,P＜0.01);骨肉瘤MVD均值为71.5(28～152),高MVD组survivin表达率显著高于低MVD组(P＜0.01).骨肉瘤中survivin mRNA、VEGF表达及MVD与组织学分级无关,与组织学分型(WHO分型)和转移有关.结论survivin基因选择性高表达于骨肉瘤组织,通过抑制细胞凋亡,参与骨肉瘤的发生发展;其对骨肉瘤血管生成和转移的作用可能与VEGF存在密切相关;survivin可能是评价骨肉瘤预后的重要参考指标.

Study on expression and correlation of survivin and VEGF in osteosarcoma

Objective To investigate the effect and relationship of survivin and VEGF on angiogenesis and metastasis of osteosarcoma by detecting the two factors expression. Methods 38 osteosarcoma cases were involved, in situ hybridization was used for the detection of survivin mRNA expression; immunohistochemistry for VEGF and CD34 protein expression; microvessel density (MVD) was counted; and then comparison with the major clinical pathological parameters of osteosarcoma was conducted. Results The expression rate of survivin mRNA and VEGF protein in osteosarcoma were all 65.8%, two factors expression had positive correlation (r = 0.546, P <0.01). The mean value of MVD of osteosarcoma was 71.5(28~152), survivin expression rate in high MVD group was significantly higher than that of the low MVD group (P <0.01). The expression of survivin mRNA, VEGF and MVD had no correlation with histological grade, but significantly according to histological classification (WHO typing) and lymph node metastasis. Conclusions Survivin gene selectively high expresses in osteosarcoma. It maybe involve in the occurrence and progress of osteosarcoma by inhibition of apoptosis; it possibly has effect on the angiogenesis and metastasis of osteosarcoma with the synergetic effect of VEGF; survivin may be an important reference index in evaluation of prognosis of osteosarcoma.
