摘 要: | 目的:初步研究不同炮制时间何首乌致泻强度与化学含量相关性,并初步建立对其致泻药理数学模型。方法:采用小鼠泻下半数有效量(ED50)为评价指标,考察何首乌生品和经4个不同炮制时间处理过的何首乌制品致泻强度的差异;用高效液相色谱法测定生品及不同制品中蒽醌类化合物含量,揭示不同何首乌样品与致泻强度二者量-时-效关系,并建立不同炮制时间的何首乌致泻药效的数学模型。结果:不同炮制时间制首乌的ED50所需给药剂量分别为:2.562、4.085、11.146、14.692、21.068g/kg(生药/体重);致泻强度顺序为:生品何首乌>1h制品何首乌>2h制品何首乌>3h制品何首乌>4h制品何首乌;结合型蒽醌含量高低顺序为:生品何首乌>1h制品何首乌>2h制品何首乌>3h制品何首乌>4h制品何首乌。讨论:何首乌生品、经不同炮制时间处理后的制何首乌之间致泻强度存在较大差异;致泻强度与结合型蒽醌含量间具有量-时-效关系;将生物检测方法和数学模型化模式联系起来,更全面的保证中药质量及中医临床用药安全。

Abstract: | Objective: A preliminary study on purgative strength among different processing time of Polygonum multiflorum and chemical content correlation, the initial establishment of the mathematical model of diarrhea induced by pharmacological. Methods: The mice purgative median effective dose ( ED50 ) as the evaluation index, Polygonum multiflorum thunb and study the differences of purgative intensity between raw medicine and 4 different processing time products of processed Radix Polygoni muhiflofi ; using high performance liquid chromatography method for determining the content of antilraquinone between raw medi- cine and processed products of in Radix Polygoni multiflori, revealing the relationship of quantity-time-effect between different samples with different processing time and diarrhoea causative intensity , and establishing the mathematical model of purgative effect of Polygonum muhiflorum Thunb. Results : The required EDS0 dosage of different processing time of Polygonum muhiflo- rum respectively: 2. 562, 4. 085, 11. 146, 14. 692, 21. 068g·kg^-1( crude drug·weight-1 ) ; purgative intensity sequence was : the raw〉l h〉2h〉3h〉4h ; combined anthraquinone content in order : the raw〉 1〉2h〉3h〉4h. Conclusion: There are obvious differences of purgative strength between raw product and processed Radix Polygoni Muitiflori , it has positive relationship between purgative strength and combined anthraquinone content on quantity-time-effect; associating biological detection method with mathematical model, the quality of traditional Chinese medicine and clinical medication safety will more comprehensively guaranteed. |
