艾灸对寒凝血瘀型痛经大鼠子宫瞬时受体电位香草酸亚型1表达的影响

摘    要:目的观察艾灸神阙穴对寒凝血瘀型痛经大鼠扭体反应及子宫组织瞬时受体电位辣椒素亚型1(TRPV1)表达的影响,并探讨神阙穴肥大细胞发挥的功能。方法将50只雌性Wistar大鼠随机分为5组:对照组、模型组、艾灸组、色甘酸钠+模型组、色甘酸钠+艾灸组,每组10只。除对照组外均构建寒凝血瘀型痛经模型,造模后给予相应治疗。观察评估各组大鼠扭体反应,并采用免疫荧光、Western blot、qRT-PCR技术,检测各组大鼠子宫组织TRPV1在蛋白和mRNA水平的表达。结果艾灸神阙穴对寒凝血瘀痛经大鼠具有明显镇痛疗效;神阙穴皮下注射色甘酸钠具有一定镇痛作用;模型组大鼠子宫TRPV1表达较对照组升高(P<0.01);艾灸可下调痛经大鼠子宫TRPV1高表达(P<0.01)而达到镇痛疗效;色甘酸钠阻断神阙穴肥大细胞使艾灸镇痛效应及对子宫TRPV1表达的调节作用不同程度地削弱。结论艾灸神阙穴通过下调痛经大鼠子宫TRPV1高表达而达到治疗作用;神阙穴区MC在此效应过程中发挥了重要作用。

Abstract:Objective To observe the umbilical moxibustion effect on transient receptor potential vanilloid 1 in uterus of rats with dysmenorrhea of cold coagulation and blood stasis tpye and explain the mast cellseffect.Methods Rats were randomly divided into 5 groups:control group,model group,moxibustion group,SCG+model group,SCG+ moxibustion group.Except for control group,the dysmenorrhea model of cold coagulation and blood stasis type was constructed.After corresponding treatment was given,Immunofluorescence,Western blot and qRT-PCR methods were used to test the transient receptor potential vanilloid 1(TRPV1) expression in ratsuterine.Results Moxibustion had obvious analgesic effect on dysmenorrhea of cold coagulation and blood stasis type. SCG had certain analgesic effect. The expression of TRPV1 in the uterus of model rats was higher than that of the control group(P<0.01). The moxibustion on navel can reduce the high expression of TRPV1 in the uterus of the dysmenorrhea rats(P<0.01) and reach the analgesic effect. SCG blocked the mast cells to weaken the analgesic effect of moxibustion on the umbilicus and its regulation on the expression of TRPV1 in the uterus.Conclusion Moxibustion on navel can achieve a therapeutic effect by down-regulating the high expression of TRPV1 in the uterus of dysmenorrhea ratsand MC plays an important role in this process.
