鱼王浆对免疫功能低下模型小鼠相关免疫细胞因子影响的实验研究

摘    要:目的研究鱼王浆对环磷酰胺致免疫功能低下模型小鼠相关免疫细胞因子的影响及量效关系。方法通过腹腔环磷酰胺(Cy)强化注射制造免疫功能低下模型小鼠,模型小鼠随机分为鱼王浆高、中、低剂量组及模型组,另设1组为空白对照组。观察比较鱼王浆高、中、低剂量对模型小鼠相关免疫因子的影响,检测外周血CD3^+、CD4^+,肿瘤坏死因子-α(TNF-α)及干扰素-γ(IFN-γ)含量,脾脏组织TNF-α蛋白表达的变化。结果鱼王浆高、中剂量组CD3^+、CD4^+的含量明显高于模型组(P〈0.01),鱼王浆低剂量组CD3^+的含量与模型组比较有所升高(P〈0.05);鱼王浆各剂量组TNF-α、IFN-γ的含量明显高于模型组(P〈0.01);鱼王浆各剂量组脾组织中TNF-α蛋白的表达明显高于模型组(P〈0.01)。结论鱼王浆能明显提高免疫功能低下模型小鼠外周血中相关免疫细胞因子CD3^+、CD4^+及TNF-α、IFN-γ的含量,增强免疫组织器官中TNF-α的表达,并且其作用具有明显的量效关系。

Abstract:Objective To study the impacts of yuwang syrup on the relevant immune cell factors and the dose-effect relationship in the hypoimmunity model mice induced by cyclophosphamide.Methods The intensified peritoneal injection of cyclophosphamide was used to prepare the hypoimmunity model mice.The model mice were randomized into a high-dose yuwang syrup group,a middle-dose yuwang syrup group,a low-dose yuwang syrup groups and a model group.One blank control group was set up additionally.The impacts of yuwang syrup of the high dose,middle dose and low dose on the relevant immune factors were observed in the model mice separately.The contents of peripheral blood CD3^+,CD4^+,tumor necrosis factor-α(TNF-α)and interferon-γ(IFN-γ)were detected.The changes in TNF-α protein expression in spleen tissue were observed.Results The contents of CD3^+ and CD4^+ in the high-dose and middle-dose yuwang syrup groups were higher apparently than those in the model group(P〈0.01).The content of CD3^+ in the low-dose yuwang syrup group was increased as compared with the model group(P〈0.05).The contents of TNF-α and IFN-γ in each yuwang syrup group were higher apparently than those in the model group(P〈0.01).The TNF-α protein expression in spleen tissue in each yuwang syrup group was higher than that in the model group(P〈0.01).Conclusion Yuwang syrup improves apparently the contents of the relevant immune cell factors such as CD3^+,CD4^+,TNF-α and IFN-γ in the peripheral blood in the hypoimmunity model mice and strengthens the expression of TNF-α in the immune tissue organ.Additionally,the dose-effect relationship is obtained obviously.
