丹蒌片对短暂心肌缺血再灌注诱导的心律失常模型大鼠的保护作用及机制

摘    要:目的探讨丹蒌片对短暂心肌缺血再灌注诱导的心律失常模型大鼠的保护作用及机制。方法 将45只健康成年Wistar大鼠随机分为假手术组、模型组、丹蒌片组,每组15只。假手术及模型组以O.1 mL/(kg·d)蒸馏水灌胃,丹蒌片组以0.53 g/mL浓度的丹蒌片混悬液0.1 mL/(kg·d)灌胃,连续给药10天,末次灌胃1 h后进行心肌缺血再灌注实验。描记结扎前、缺血和再灌注时心电图,全程监测各组大鼠颈动脉血压。观察各组大鼠心肌缺血再灌注不同时间ST段变化,再灌注30 min内大鼠心室颤动、室性早搏、室性心动过速的发生次数及持续时间,并检测心肌组织匀浆Na^+-K^+-ATP酶及Ca^(2+)-ATP酶的活性。结果丹蒌片组大鼠颈动脉血压及心率略低于模型组,两组比较,差异无统计学意义(P〉0.05)。与假手术组比较,模型组缺血0、5、7 min ST段明显抬高(P〈0.05);与模型组比较,丹蒌片组ST段抬高程度显著降低(P〈0.01);模型组缺血5 min时ST段较0 min时进一步抬高,但丹蒌片组抬高程度仍明显低于模型组(P〈0.05);两组缺血7 min时ST段抬高程度比较,差异无统计学意义(P〉0.05)。再灌注0 min时,模型组及丹蒌片组ST下降超过缺血时1/2,两组大鼠心肌缺血程度比较,差异无统计学意义(P〉0.05)。与模型组比较,丹蒌片组致命性及非致命性室颤发生率、室性心动过速、室性早搏的发生频率及持续时间明显减少(P〈0.05),Na^+-K^+-ATP酶、Ca^2+-ATP酶活性增加(P〈0.05)。结论丹蒌片对短暂心肌缺血再灌注诱导的心律失常有显著的保护作用,其作用可能与降低心肌缺血程度,增加离子转运通道相关酶活性有关。

Effect of Danlou Tablet on Arrhythmia Model Rats Induced by Transient Myocardial Ischemia/Reperfusion

Abstract:Objective To explore the effect of Danlou Tablet(DT) on arrhythmia model rats induced by transient myocardial ischemia/reperfusion(l/R).Methods Totally 45 healthy Wistar rats were randomly divided into 3 groups,the sham-operation group,the model group,and the DT group,15 in each group.Rats in the sham-operation group and the model group were administered with distilled water by gastrogavage at the daily dose of 0.1 mL/kg.Rats in the DT group was administered with 0.53 g/mL DT suspension by gastrogavage at the daily dose of 0.1 mL/kg.All medication was lasted for 10 successive days.The myocardial l/R experiment was performed at 1 h after the last gastrogavage.ECG was performed before ligation and at l/R.The jugular arterial blood pressure of all rats was measured during the whole course.ST segment changes were observed at each time point of l/R.The ventricular fibrillation,the premature ventricular,the number and the duration of ventricular tachycardia within 30 min reperfusion were also observed.Activities of Na^+-K^+ATPase and Ca^(2+) ATPase in the myocardium homogenate were detected as well.Results The jugular arterial blood pressure and the heart rate were slightly lower in the DT group than in the model group,but with no statistical difference(P〉0.05).Compared with the sham-operation group,the degree of ST segment was obviously elevated in the model group at 0,5,and7 min(P〈0.05).It was significantly lower in the DT group than in the model group(P〈0.01).ST segment was more elevated at 5 min than at 0 min in the model group,but the degree of ST segment elevation was still obviously lower in the DT group than in the model group(P〈0.05).There was no statistical difference in the degree of ST segment elevation at 7 min between the two groups(P〉0.05).At 0 min when the decrement of ST segment exceeded one half the ischemia,there was no statistical difference in the degree of myocardial ischemia between the model group and the DT group(P〈0.05).Compared with the mo
