| 摘 要： | 目的：探讨周围神经损伤后脊髓神经元退变与组织型纤溶酶原激活物（tPA）及其抑制物1（PAI-1）、神经丝氨酸蛋白酶抑制剂（NSP）表达的关系。方法：雄性SD大鼠（56只）随机分为实验组（50只）和对照组（6只。无手术）。实验组行坐骨神经离断术（SCNT）,于术后1、4、7、14和21 d取伤侧脊髓L4~6节段,观察前角神经元形态学改变及tPA、PAI-1和NSP表达。结果：与对照组比,实验组L4~6tPA、NSP表达增加,NSP在1 d内达峰值后逐渐下调,tPA水平则在7 d升达最高峰,此时前角外侧核神经元存活率开始减少（P〈0.01）。术后21 d,tPA仍未恢复正常水平（P〈0.05）,脊髓前角运动神经元存活率61.6%。两组脊髓均未测到PAI-1表达。结论：SCNT后L4~6前角神经元退变,可能与损伤刺激脊髓灰质内神经元和小胶质细胞合成、释放tPA增加有关,同时,损伤也促使tPA抑制物NSP表达上调,后者可能发挥神经保护作用。
