周围神经损伤后脊髓神经元退变与组织型纤溶酶原激活物及其抑制物表达的关系研究

摘    要:目的:探讨周围神经损伤后脊髓神经元退变与组织型纤溶酶原激活物(tPA)及其抑制物1(PAI-1)、神经丝氨酸蛋白酶抑制剂(NSP)表达的关系。方法:雄性SD大鼠(56只)随机分为实验组(50只)和对照组(6只。无手术)。实验组行坐骨神经离断术(SCNT),于术后1、4、7、14和21 d取伤侧脊髓L4~6节段,观察前角神经元形态学改变及tPA、PAI-1和NSP表达。结果:与对照组比,实验组L4~6tPA、NSP表达增加,NSP在1 d内达峰值后逐渐下调,tPA水平则在7 d升达最高峰,此时前角外侧核神经元存活率开始减少(P〈0.01)。术后21 d,tPA仍未恢复正常水平(P〈0.05),脊髓前角运动神经元存活率61.6%。两组脊髓均未测到PAI-1表达。结论:SCNT后L4~6前角神经元退变,可能与损伤刺激脊髓灰质内神经元和小胶质细胞合成、释放tPA增加有关,同时,损伤也促使tPA抑制物NSP表达上调,后者可能发挥神经保护作用。
