抗肿瘤药物与维拉帕米联合对KBV200细胞P-糖蛋白表达的影响研究

摘    要:目的:探讨6种常用抗肿瘤药物长春新碱、阿霉素、平阳霉素、顺铂、5-Fu、环磷酰胺与维拉帕米联合对KBV200细胞P-糖蛋白表达的影响.方法:以喉癌KBV200细胞株为研究对象,通过流式细胞术检测6种抗肿瘤药物作用KBV200细胞的P-糖蛋白表达以及联合维拉帕米对P-糖蛋白表达的影响.结果:维拉帕米分别与长春新碱、阿霉素、平阳霉素、环磷酰胺联合作用,使KBV200细胞的P-糖蛋白表达下降80.17%、89.69%、19.19%及53.48%.其中维拉帕米与长春新碱、阿霉素作用较强.结论:维拉帕米分别与长春新碱、阿霉素、平阳霉素、环磷酰胺联合作用后,P-糖蛋白的表达水平明显下降,对KBV200细胞耐药性的逆转有一定作用.

Abstract:Objective: To observe the effect of verapamil and 6 chemotherapeutic agents vincristine,adriamycin,bleomycin,cyclophosphamide,5-Fu,cisplatin on the P-glucoprotein(P-gp) of the KBV200 cell line.Methods: Flow cytometer(FCM) assay was used to investigate the expression of P-gp on the KBV200 cell line in the different conditions.Results: Verapamil act with vincristine,adriamycin,pingyangmycin and cyclophosphamide separately decreased expression of P-gp on KBV200 cell line by 80.17%,89.69%,19.19% and 53.48% Among all the combinations,the synergistic effects of varapamil act with vincristine and adriamycine show a better effect.Conclusion: Verapamil act with vincristine,adriamycin,pingyangmycin and cyclophosphamide separately can decrease expression of P-gp on KBV200 cell line significantly,and can reverse the drug resistance of KBV200 cell line.
