毛细管区带电泳法对马钱子药材中生物碱质量控制的研究

摘    要: 目的建立马钱子药材中生物碱质量控制的方法。方法利用毛细管区带电泳法,以来涂层石英毛细管(75μm×50 cm)为分离通道,80mmol·L-1Tris-H3BO3(醋酸调pH至4.0)为运行缓冲溶液,分离电压23 kV,进样3.45×103 Pa,10s;柱温25℃;DAD检测波长214 nm。结果士的宁和马钱子碱的浓度均在1.0~120 mg·L-1内呈良好线性关系,r分别为0.9999和0.9997;加样回收率分别为96.57%(RSD=1.73%)和102.04%(RSD=1.62%);最低检测限分别为0.490 mg·L-1和0.957 mg·L-1。结论实验证明利用毛细管区带电泳法,马钱子药材中士的宁和马钱子碱能够与其他组分得到很好的分离,方法简单、快速,可作为中药材中有效成分质量控制的方法。

Abstract:OBJECTIVE To establish the methed for the quality control of alkaloid in Strychnos nux-vomica L.METHODS The sepa- ration was performed on a fused-silica capillary of 75μm×50 cm using the method of capillary zone electrophoresis.80 mmol·L-1 Tris-H3BO3 (pH 4.0)was used as running buffer,The samples were injected with the pressure at 3.45×103 Pa for 10 s,separated with the voltage of 23 kV at the temperature 0f 25℃and determined at the wavelen-gth of 214 nm.RESULTS The calibration curve showed a good linearity within the range of 1.0~120 mg·L-1(r=0.999 9)for strychnine and 1.0-1120 mg·L-1(r=0.999 7)for brucine.The recovery were 96.57% (RSD=96.57%)and 102.04%(RSD=1.62%)and the lowest limits of detection were 0.490 and 0.957 mg·L-1 for strychnine and brucine respectively.CONCLUSION By means of this methed,strychnine and brucine can be well separated from the other components. The method is convenient and rapid,and can be used to control the quality of effective components in drugs.
