口服枸杞多糖对实验性动脉粥样硬化斑块预防作用的影响

摘    要:目的:观察口服枸杞多糖(LBP)对动脉粥样硬化(AS)的预防作用。方法:40只雄性新西兰白兔随机分成5组:正常组、模型组、高剂量LBP组、中剂量LBP组、低剂量LBP组,每组8只。正常组始终喂饲普通饲料;模型组喂普通饲料+质量分数为2%胆固醇,高剂量LBP组、中剂量LBP组及低剂量LBP组分别喂饲普通饲料+质量分数为2%胆固醇+20mg/kgLBP(高剂量组)、10mg/kgLBP(中剂量组)和5mg/kgLBP(低剂量组)。在第4、8、12周末,分别测量总胆固醇(Tch)、甘油三脂(TG)、高密度脂蛋白胆固醇(HDL—C)、低密度脂蛋白胆固醇(LDL—C)、丙二醛(MDA)、超氧化物歧化酶(SOD)。第12周一次处死所有动物并观察主动脉形态学变化及应用免疫组织化学染色进行指标的检测及分析。结果:与正常组相比,模型纽Tch、TG、HDL-C、LDL—C、MDA明显升高(P〈0.01),SOD活力降低(P〈0.01);三组LBP组与模型组比较,Tch、TG、LDL—C、MDA明显降低(P〈0.01),HDL-C明显升高(P〈0.01),SOD活力升高(P〈0.01);而高、中、低剂量LBP组比较无明显差异。通过免疫组织化学染色发现:LBP组与模型组比较,主动脉斑块面积和斑块内平滑肌细胞明显减少;BCL-2,MMP-13的表达降低。结论:LBP不仅能够降脂,它还可以减少BCL-2,MMP-13的表达,这说明LBP能够对AS斑块性质产生影响,由此阻止动脉粥样硬化的发生发展。

EFFECTS OF THE ORAL LYCIUM BARBARUM POLYSACCHARIDE ON THE PREVENTION OF EXPERIMENT ATHEROSCLEROSIS

Abstract:Objective: To investigate the effects of Lycium Barbarum Polysaccharide (LBP) on the rabbit atherosclerotic (AS) plaque formation. Methods: Forty healthy male New Zealand rabbits were randomized into 5 groups : normal group, atherosclerosis model group, and high, medium and low LBP groups. There were 8 rabbits in each group. Normal group always fed with normal diet, atherosclerosis model group fed with normal diet and 2% cholesterol, LBP groups fed with normal diet, 2% cholesterol and 20 mg/kg, 10mg/kg or 5mg/kg LBP. At the ends of 4,8,12 weeks, total cholesterol (Tch), triglyceride (TG), high density lipoprotein cholesterol ( HDL - C ), low density lipoprotein cholesterol ( LDL - C ), Malonaldehyde ( MAD ) and superoxide dismutase ( SOD ) were measured. At the end of the 12 weeks, all rabbits were killed. The morphological change of aorta was observed and further quantified using immunohistochemistry result. Results:Compared with normal group, the model group's Tch,TG,HDL- C ,LDL- C ,MDA increased. Compared with model group, the LBP groups' Tch, TG, LDL - L, MDA lowered, HDL - C, SOD increased, but the significant changes have not happened in the LBP groups. As found with SABC immunohistochemistry, Bcl -2 and MMP - 13 were localized in AS plaques. Compared with model group, the LBP groups showed significantly lower level of Bcl -2 and MMP - 13 expression. Conclusion:In addition to reduction of blood lipids, LBP appeared to markedly reduce the expression of Bcl -2 and MMP - 13 in AS plaques. It underlies its effeetion on the plaque quality and by which LBP can prevent the development of AS.
