摘    要:目的:建立茯苓中茯苓酸的含量测定方法,比较12个不同产地茯苓中有效成分茯苓酸的含量,为评价不同产地茯苓的品质提供参考依据。方法:采用反相高效液相色谱蒸发光散射法(HPLC-ELSD),Kromasil 100-5C18色谱柱(250mm×4.6mm,5μm),柱温30℃,流动相为乙腈-0.2%甲酸(78:22),流速1.0ml·min-1,蒸发光检测器漂移管温度:40℃,压力:3.5bar。结果:茯苓酸在0.488~2.440μg范围内线性关系良好(r=0.999 8),回收率试验平均为100.05%,RSD=0.47%;12个不同产地茯苓中茯苓酸的含量差异较大。结论:该方法简便、稳定、专属、可靠性强,可用于比较不同产地茯苓有效成分茯苓酸的含量,能有效评价各地茯苓药材的质量。

Abstract:Objective: To establish a method for the determination of pachymic acid in Poria cocos from twelve different areas in order to provide a basis to evaluate the quality of Poria cocos from different habitats. Method : HPLC-ELSD method was used on a Kro- masil 100-5C18 column (250 mm ×4. 6 mm,5 μm) with acetonitrile-0. 2% methanoie acid (78: 22) as the mobile phase at a flow rate of 1.0 ml. rain-1. The column temperature was 30 ℃, the temperature of drift tube for ELSD was 40 ℃ and the pressure was 3.5 bar. Result: Paehymic acid showed a fine linear relationship with the peak areas between 0. 488 and 2. 440 μg ( r = 0. 999 8 ). The average recovery was 100. 05% with RSD of 0. 47%. There was significant difference in the content of paehymic acid in Poria cocos from differ- ent areas. Conclusion: The method is proved to be convenient, stable, exclusive and feasible for the quality assessment of pachymie acid, which is the main effective component in Poria cocos. The method also can evaluate the quality of Poria cocos from different habi- tats effectively.
