应用TBA荧光法检测了兔眼单纯晶体囊外摘除后和晶体囊外摘除并后房型人工晶体置入后房水内丙二醛含量,这二种手术后房水内丙二醛含量比正常眼显著增高(P<0.005,P<0.001)。结果提示前房内发生了脂质过氧化反应,这一发现有利于对眼内后房型人工晶体表面细胞反应及前膜形成进一步认识。  相似文献

白内障手术结膜囊及房水细菌培养的临床观察   总被引:3,自引:0,他引:3  
目的 探讨白内障术中结膜囊、房水污染与眼内炎的关系。方法 对 74例 80眼白内障囊外摘除术中结膜囊及房水进行细菌培养。结果  16 0份细菌培养标本中 ,细菌培养阳性者 18株 ,平均培养阳性率为 11.3%。房水为 8株 ,检出率为 10 .0 % ;结膜囊为 10株 ,检出率为 12 .5 %。以表皮葡萄球菌为主 (11 18) ,其中 3眼结膜囊及房水检出同一菌株。检出阳性的 15眼无 1例发展成眼内炎。结论 白内障摘除术后前房有少量细菌 ,虽不致发生眼内炎 ,但不可掉以轻心。  相似文献

Objective To observe effects of penetrating cornea injury combined with seawater immersion on the levels of aqueous humor TNF-α in rabbits. Methods 10 adult healthy grey rabbits were used for the experiment. The rabbit eye model of comea blast injury was developed by using a single firecracker. For each rabbit, the right eye was the experimental eye (seawater was injected into the anterior chamber through the cornea incision, simulating seawater immersion) and the left eye was the control (treated with 0.9% sodium chloride). Levels of aqueous humor TNF-α were detected by using double-antibody sandwich ELISA on the 1st, 2nd, yrd, 4th, 5th, and 7th days following seawater immersion or 0.9% sodium chloride injection. Results Following development of the model, levels of aqueous humor TNF-α in the experimental eye on the 1st, 2nd, 3rd days were 190.338, 156. 107,117.018 ng/L respectively, and significant differences could be seen when comparisous were made between the experimental and control eyes. The level of aqueous humor TNF-α on the 1st day was the highest and obviously reached the peak. Conclusions Level of aqueous humor TNF-α in the experimental eye with penetrating cornea injury combined with seawater immersion was significantly higher than that of the control eye, indicating that TNF-α was clearly involved in the pathological change and it played an important role in the process.  相似文献

Problems associated with the use of 5-iodo-2-deoxyundine (IDU) in the treatment of herpes simplex keratitis can be attributed largely to the polar nature of IDU resulting in its poor permeability across the lipoidal epithelial layer of the corneal membrane. Five aliphatic 5-esters of IDU were synthesized and evaluated as prodrugs for potential use in the treatment of deep ocular infections such as stromal keratitis, iritis, and even retinitis. A parabolic relationship between in vitro corneal membrane permeability and carbon chain length of prodrugs is evident. For a given prodrug, enzymatic hydrolysis proceeded most readily in iris–ciliary body, followed by cornea and aqueous humor. An increase in carbon chain length made the prodrugs more enzymatically labile but more resistant to chemical hydrolysis at pH 7.4 and 34°C. The 5-butyryl ester of IDU exhibited an approximately fourfold increase in aqueous humor IDU concentration relative to IDU at 25 min following instillation of 25-µl 5 mM solutions.  相似文献

The aim of this study was to examine the kinetic profile of bioactive TNF levels in aqueous humour of rabbit eyes undergoing corneal allograft rejection and to investigate the effect of locally blocking TNF activity after corneal transplantation. In a rabbit corneal transplantation, endothelial allograft rejection was identified and correlated with increase in central graft thickness. Samples of aqueous humour obtained on alternate days following transplantation were tested for TNF mRNA and bioactive TNF protein. To investigate the effect of locally blocking TNF activity in allograft recipients, the fusion protein TNFR-Ig was administered by injections into the anterior chamber after transplantation. Pulsatile increases in levels of this cytokine were found in 14 of 15 allograft recipients. Peaks of TNF bioactivity preceded by varying intervals the observed onset of rejection in allograft recipients. TNF levels were not elevated in aqueous humour from corneal autograft recipient controls or in serum of allografted animals. mRNA levels were elevated before onset of and during clinically observed allograft rejection. In three of seven animals receiving TNFR-Ig injections on alternate days from day 8 to day 16 post-transplant, clear prolongation of corneal allograft survival was demonstrated. Bioactive TNF is present in aqueous humour following rabbit corneal allotransplantation. Rather than correlating directly with endothelial rejection onset, pulsatile peak levels of TNF precede and follow the observed onset of endothelial rejection. Blockade of TNF activity prolongs corneal allograft survival in some animals, indicating that this cytokine may be a suitable target in local therapy of corneal allograft rejection.  相似文献

The differences in flurbiprofen disposition in the aqueous humor and the plasma were examined after systemic doses. Steady state plasma concentrations of flurbiprofen (20–60 g/mL) were achieved via intravenous infusion to albino rabbits. Flurbiprofen demonstrated linear systemic kinetics throughout the dosing range, with constant body clearance and unbound fraction in plasma. At steady state, aqueous humor drug concentrations depended on the corresponding plasma drug concentration. Two clearance terms—CLso, the systemic clearance to ocular tissues, and CLos, the ocular clearance to systemic circulation—were used. After systemic doses, the drug concentration in the aqueous humor was related to that in the plasma as well as to the ratio of these two clearances. Flurbiprofen was extensively bound to plasma proteins and showed limited ocular distribution; its CLso to CLso tratio was very small. Thus, the concentration of flurbiprofen in the aqueous humor after systemic doses was lower than that obtained after ophthalmic doses. A plasmapheresis technique was utilized to lower the plasma protein concentrations to 60% of normal levels. As a consequence, flurbiprofen demonstrated reduced aqueous humor protein concentrations, increased unbound fractions in the plasma and the aqueous humor, elevated aqueous humor drug concentrations, and elevated total body clearance. The unbound body clearance stayed unchanged. Our study indicated that a drug should present a significant CLso/CLos ratio in order to achieve therapeutic concentrations in the eye via systemic doses. The drug-protein binding kinetics can be different between the plasma and the aqueous humor circulations. Because the ocular compariment is very small compared to the overall systemic distribution of flurbiprofen, it has little effect on the steady state systemic concentrations.  相似文献

目的:探讨前部增生性玻璃体视网膜病变(anterior prolieferative vitreoretinopathy,aPVR)低眼压状态下房水生成率变化,从房水动力学的角度揭示aPVR引起慢性低眼压的发病机制。方法:利用培养的同种兔皮肤成纤维细胞制作aPVR引起慢性低眼的动物模型。于术前及术后不同时间点分别观测眼压、术后14d,28d,56d分别以高效液相色谱仪测量房水荧光素清除率,进一步计算房水生成率。结果:术后2周、4周、8周实验组平均眼压、房水生成率明显低于对照组(P<0.05或P<0.01)。结论:在aPVR病理状态下,低眼压的形成与房水生成率下降有关。  相似文献

对115例原发性闭角型青光眼和34例老年性白内障患者的房水蛋白进行了测定。结果:闭角型青光眼 疾病分类各组及中医证型各组均表现为房水白蛋白和总蛋白含量增高,与老年性白内障组相比,各组均有非常显著性差异(P<0.01)。闭角型青光眼疾病分类组间比较及中医证型组间比较,房水白蛋白和总蛋白含量亦均有显著性差异(P<0.05)或非常显著性差异(P<0.01)。证明原发性闭角型青光眼患者的血-房水障碍功能遭受破坏,房水的粘度增加,房水淤积于眼内。  相似文献
