Summary We present a simple method for the isolation of DNA from agarose gels that is economic, fast, and independent of electrical equipment. DNA fragments of up to 6 kb can be easily extracted within 5 min using a disposable plastic syringe and filter paper. Total extraction of DNA fragments between 10 and 20 kb in size is achieved by concentrating the DNA flushed from the gel in a DNA-binding column. 相似文献

Novel polymer electrolyte materials based on a polyelectrolyte‐in‐ionic‐liquid principle are described. A combination of a lithium 2‐acrylamido‐2‐methyl‐1‐propanesulfonic acid (AMPSLi) and N,N′‐dimethylacrylamide (DMMA) are miscible with the ionic liquid, 1‐ethyl‐3‐methylimidazolium dicyanamide (EMIDCA). EMIDCA has remarkably high conductivity (≥ 2 · 10?2 S · cm?1) at room temperature and acts as a good solvating medium for the polyelectrolyte. At compositions of AMPSLi less than or equal to 75 mol‐% in the copolymer (P(AMPSLi‐co‐DMAA)), the polyelectrolytes in EMIDCA are homogeneous, flexible elastomeric gel materials at 10 ? 15 wt.‐% of total polyelectrolyte. Conductivities higher than 8 · 10?3 S · cm?1 at 30 °C have been achieved. The effects of the monomer composition, polyelectrolyte concentration, temperature and lithium concentration on the ionic conductivity have been studied using thermal and conductivity analysis, and pulsed field gradient nuclear magnetic resonance techniques.

New types of hydrophilic gels based on N-(2-hydroxypropyl)methacrylamide which contain oligopeptide sequences in the crosslinks were prepared. These gels are enzymatically degradable by chymotrypsin. The rate of their degradation may be varied within a broad range by changes in the length and detailestructure of the oligopeptide sequence in the crosslinks and by changing their network density. 相似文献

Structural-mechanical strength and swelling of gels consisting of gelatin and potassium hyaluronate (PHY) or potassium protine-chondroitin-4-sulfate (PPCS) were shown to depend on the ratio between the components. With low concentrations of PHY and PPCS the minimum of structural-mechanical strength coincided with the maximum of swelling of the gels. In zones of neutralization of the positive electric charges of gelatin by macropolyanions, high structural-mechanical strength of the gels coincided with the minimum of swelling. In high concentrations of PHY, structural-mechanical strength and swelling of the gel became equal to the values characteristic of a gel consisting of gelatin only, but in high concentration of PPCS there was an additional parallel increase in this strength and in swelling of the gel.(Presented by Academician of the Academy of Medical Sciences of the USSR S. Summary: The equilibrium swelling degree, modulus of elasticity and the spatial inhomogeneity of poly(N,N‐dimethylacrylamide) (PDMAAm) hydrogels were investigated over the entire range of the initial monomer concentration. The degree of dilution of the networks after their preparation was denoted by ν, the volume fraction of crosslinked polymer after the gel preparation. The linear swelling ratio of the gels increased linearly with increasing ν. Depending on the value of ν, three different gel regimes were observed: (1) For ν < 0.3, increasing ν decreases the extent of cyclization during crosslinking so that the effective crosslink density of gels increases with rising ν. (2) For 0.3 < ν < 0.7, increasing ν reduces the accessibility of the pendant vinyl groups during crosslinking due to steric hindrance at high polymer concentrations. As a result, the effective crosslink density of gels decreases with increasing ν. (3) For ν > 0.7, the modulus of elasticity increases sharply with increasing ν due to the increasing extent of chain entanglements in this high concentration regime. Static light scattering measurements on the gels show that the degree of spatial gel inhomogeneity in PDMAAm gels attains a maximum value at ν = 0.06. The appearance of a maximum as well as the ν‐dependence of scattered light intensities from gels was successfully reproduced by the theory proposed by Panyukov and Rabin.

利用二氧化硅特异地吸附核素的特性,建立了从琼脂糖凝胶中回收DNA的方法。结果表明本方法对1300 ̄3500bp的DNA回收效果较好,回收效率达60% ̄70%,100 ̄1500bp,3500 ̄5000bp的DNA亦能被有效地回收。通过对回收DNA的酶切,连接等实验,证实了所回收的DNA片段能够满足进一步的实验要求。该方法简单,快捷,经济,适用,值得推广。 相似文献

The influence of electrolytes on the retention of organic cationic solutes in reversed-phase high-performance liquid chromatography (RP-HPLC) has been investigated. The effects of the nature and concentration of electrolytes and mobile phase pH on the retention of two model quaternary ammonium compounds were studied on mu-Bondapak C18 stationary phase with aqueous methanolic eluents. The nature and concentration of inorganic cations added to the mobile phase modified the retention of the solutes. The counter anion of the added electrolyte did not perceptibly influence solute retention at constant mobile phase pH, although it did significantly influence solute retention when the electrolytes were added to unbuffered mobile phases. The retention data are consistent with the inclusion of an ion exchange contribution to the retention of cationic solutes in the systems investigated. 相似文献
