Isolated bovine articular cartilage chondrocytes and intact slices of cartilage were investigated to determine the effects of low-temperature cryopreservation on articular cartilage. Studies have focused on prefreezing conditions of cartilage, including the incubation medium and temperature of incubation, type and toxicity of the cryopreservative used, and the penetration of cryopreservative agents into cartilage cells. Cartilage freezing conditions were examined with respect to rate of freezing, controlled differential freezing rates, the ultimate storage temperature, and the time of storage. Cartilage thawing conditions were observed to ascertain the role of membrane osmotic stress during thawing and the effect of variable thawing rates on the viability of chondrocytes. Careful control of these variables can yield cartilage with cellular viability of over 50%. Optimum cryopreservation of viable cartilage should include prefreezing treatment with 7.5%-10% DMSO in nutrient medium, controlled slow freezing to -70 degrees, and rapid thawing in DMSO containing medium. A significant number of chondrocytes in deep-frozen cryopreserved articular cartilage can survive. The work recommends continued clinical use of deep-frozen cartilage.  相似文献

Background

To evaluate the safety, tolerability, pharmacokinetics, and dose-limiting toxicities of a single intravitreal (IVT) injection of PF-04523655, a 19-nucleotide, O-methyl stabilized, double-stranded small interfering ribonucleic acid targeting the RTP801 gene in patients with neovascular age-related macular degeneration (AMD).

Methods

Prospective, phase 1, clinical multicentre trial, enrolled 27 patients with neovascular AMD unresponsive to prior treatment and best corrected visual acuity (BCVA) ≤20/200 in the study eye in stratum 1: (dose-escalating, open-label: 50 to 3000 μg of PF-04523655) and 27 patients who had potential to benefit from therapy and BCVA of ≤20/100 and ≥20/800 in stratum 2 (parallel, masked study of 1000, 1500, 2250, and 3000 μg of PF-04523655). The primary outcome was safety and tolerability assessment as well as pharmacokinetic profiling following a single IVT injection of PF-04523655.

Results

Doses of PF-04523655 ≥400 μg were generally detectable in the plasma at 1, 4, and 24 h post-injection. And all doses were below the lowest level of quantification by day 14. A single IVT injection of 50 to 3000 μg of PF-045237655 was generally safe and well tolerated over 24 months. There were no dose-limiting toxicities.

Conclusion

A single IVT injection of PF-0523655 ≤3000 μg seems safe and well tolerated in eyes with neovascular AMD.  相似文献

OBJECTIVE: To describe the properties of a semistructured research interview of parents designed to evaluate attention-deficit hyperactivity disorder (ADHD) and related psychopathology. METHOD: We examined interrater reliability in 48 videotaped interviews randomly selected from a large clinic sample. We examined convergence of the Parent Interview for Child Symptoms (PICS) and Conners' Parent Rating Scale (CPRS) scores in 594 clinic-referred cases and 26 control subjects, comparing the groups generated by cross-tabulation on measures of intelligence, academic achievement, and inhibitory control. RESULTS: Intraclass correlation coefficients for symptom scores of ADHD, oppositional defiant disorder (ODD), and conduct disorder (CD) were excellent. We found good reliability for diagnoses of ADHD (e = 0.73) and CD (kappa = 0.73) and excellent reliability for the diagnosis of ODD (kappa = 0.80). Two-thirds of cases were classified similarly on the PICS and the CPRS. Greater impairment in inhibitory control was observed in cases identified as ADHD by the PICS, compared with those identified by the CPRS-ADHD index. CONCLUSIONS: The PICS displays good reliability and validity, indicating that a balance between clinical applicability and reliability can be achieved in the development of clinical research instruments.  相似文献
