目的:研究硒化壳聚糖对人早幼粒细胞性白血病HL60细胞分化的影响,并探讨该影响与细胞c-myc和c-fos蛋白表达的关系。方法:取HL60细胞分为5组,分别为25、50、100mg·L-1硒化壳聚糖组、空白对照组(培养液)和阳性对照组(二甲基亚砜)。各组经相应试药作用后,采用硝基蓝四氮唑(NBT)还原法检测HL60细胞分化情况,观察NBT阳性细胞数;流式细胞术检测c-myc和c-fos蛋白表达。结果:与空白对照组比较,硒化壳聚糖各组作用与阳性对照组相似,NBT还原阳性细胞明显增多,c-myc蛋白表达显著下降(P<0.01或P<0.05),c-fos蛋白表达显著升高(P<0.01或P<0.05)。结论:硒化壳聚糖可能通过增强HL60细胞c-fos表达及下调c-myc表达来诱导细胞分化。  相似文献

The nucleus accumbens(NAc) plays an important role in reward,pleasure,laughter,addiction,aggression,fear,and the placebo effect,however,its role in sleep-wake regulation remains unclear.Dopamine D1 and D2 receptor(R) and adenosine A1R and A2AR are densely localized in the core and shell of NAc,and have been reported to be critically involved in sleep-wake regulation,we hypothesized that NAc might also play an important role in the regulation of sleep-wake cycle.In the present study,we made cell body-specific lesions either in the core or shell of NAc by ibotenic acid,and observed the arousal effects of modafinil and caffeine,which have been reported to be mediated mainly by dopamine receptors and adenosine A2AR,respectively.Rats with NAc core lesions exhibited a 26.5% increase in wakefulness,a weaker response in terms of sleep rebound after a 6-h sleep deprivation,and less arousal effects of modafinil or caffeine,compared with control rats.Furthermore,these lesioned rats showed a strong response to an ip saline injection,as displayed by a striking 165% prolongation in the latency to sleep.Similarly,the rats with NAc shell lesions showed a 17.4% increase in wakefulness,but the arousal effects of modafinil and caffeine in these lesioned rats were the same as that in control rats.These results indicate that both core and shell of the NAc are essential in sleep-wake regulation,with the core being more important than the shell.  相似文献

目的:研究精神分裂症遗传高危人群失匹配负波及时频特征,探索精神分裂症遗传风险识别的客观指标。方法:采用横断面设计,纳入精神分裂症患者(SCZ)、精神分裂症遗传高危人群(FHR)及健康对照(HC)各19例。采用精神病风险综合征结构式访谈(SIPS)、阳性和阴性症状量表(PANSS)及阴性症状评估量表-16(NSA-16)评估精神症状严重程度,采用精神分裂症认知功能简短评估(BACS)评估神经认知功能。利用Oddball范式采集脑电信号,比较持续时间失匹配负波(dMMN)振幅,theta诱发功率及相位一致性(ITC)。结果:SCZ的NSA-16得分高于FHR及HC(P<0.001),BACS得分低于FHR及HC(P<0.001)。SCZ的dMMN振幅低于FHR及HC(P<0.05),theta诱发功率低于FHR及HC(P<0.05)。三组间ITC差异存在趋势(P=0.078)。结论:本研究未发现精神分裂症遗传高危人群与健康人群相比存在dMMN及theta振荡异常。  相似文献
